Scientists find new target for Parkinson’s disease

Contrary to earlier discovery that a chemical found in a synthetic opioid, MPTP, induced a form of Parkinson’s disease, a new study has found that it is an enzyme in the body that can metabolise compounds formed in the brain from alkaloids causing a neurodegenerative condition in mice.

The study led by Narayan Avadhani and Mrittika Chattopadhyay, suggested that the enzyme, mitochondrial CYP2D6, presents a potentially powerful new target for Parkinson’s treatment. “Over the past two or three decades, researchers have tried inhibiting the process by they believed MPTP was metabolised, with mixed success,” said Avadhani.

“We believe that mitochondrial CYP2D6 is the more direct drug target, which might prove better in treating idiopathic Parkinson’s disease,” added Avadhani. The study published in the ‘Journal of Biological Chemistry’ investigated the mechanism of Parkinson’s disease when a specific cause cannot be pinpointed. Previous studies have shown that MPTP and similar toxic compounds induce Parkinson’s disease in rodents and primates.

The mechanism of action, as scientists understood it, involved the compounds being oxidised to form MPP+, a toxic metabolite. The enzyme that was believed to be responsible is called monoamine oxidase B (MAO-B), present in the nervous system’s glial cells. In that conception of the mechanism, MPP+ was thought to then be transferred to dopamine neurons by dopamine transporter proteins, and, indeed, Parkinson’s is characterised by unusually low dopamine levels in the brain.

Researchers have tried to stem the effects of Parkinson’s by targeting two players in this presumed pathway, both MAO-B and the dopamine transporter protein, with only mixed success. In earlier work, Avadhani and colleagues had shown that the enzyme CYP2D6, localised to the body’s energy factories, the mitochondria, could play a role in metabolising MPTP to MPP+.

In the new investigation, they took a closer look at beta-carbolines and isoquinolines, toxins that resemble MPTP which the body produces from substances found in tobacco smoke, alcohol, and some foods. They found that, instead of MAO-B, it was mitochondrial CYP2D6 that activate the beta-carbolines and isoquinolines inside the dopamine-producing neurons, rather than the glial cells.

This route of activation, in a mouse model, results in neuronal damage and oxidative stress, symptoms akin to Parkinson’s. “CYP2D6 is known to play a role in influencing the activity of a number of drugs,” said Avadhani. In an attempt to target this pathway, the researchers showed that mice lacking CYP2D6 did not exhibit severe symptoms than mice with the protein did. In addition, an inhibitor of CYP2D6 prevented neuronal damage in the mice.

“The CYP2D6 inhibitor ajmalicine is a member of the reserpine family of alkaloids, found in the plant Rauwolfia serpentine and was long used in India for treating mental illness, such as paranoia and schizophrenia,” said Avadhani. “Mitochondrial targeting of such compounds is likely to be effective in treating Parkinson’s patients, and pursuing that is our future strategy,” said Avadhani.

Lack of sleep affects fat metabolism

Healthy sleep habits can make a big difference in one's life but work week keeps us all a bit short on sleep, which can be severely harmful, says a study.

Researchers have reported that just a few days of sleep deprivation can make participants feel less full after eating and metabolise the fat in food differently.


Sleep disruption has been known to be having harmful effects on metabolism for some time.

One of the senior authors of the new study, contributed to a lot of research demonstrating that long-term sleep restriction puts people at a higher risk of obesity and diabetes.


However, he said, most of those studies have focused on glucose metabolism, which is important for diabetes, while relatively few have assessed digestion of lipids from food.


Another researcher said that after participants spent a week getting plenty of sleep at home, the 15 healthy men in their 20s checked into the sleep lab for the ten-night study.


For 5 of those nights, the participants spent no more than 5 hours in bed each night.


To find out how the uncomfortable schedule affected metabolism, the researchers gave participants a standardised high-fat dinner, a bowl of chili mac, after 4 nights of sleep restriction.


It was very palatable- none of our subjects had trouble finishing it- but very calorically dense, she said. Most participants felt less satisfied after eating the same rich meal while sleep-deprived than when they had eaten it well-rested.


Then researchers compared blood samples from the study participants. They found that sleep restriction affected the postprandial lipid response, leading to faster clearance of lipids from the blood after a meal.


This study was highly controlled, which makes it an imperfect model for the real world, she said. It focused on healthy young people, who are usually at a lower risk of cardiovascular disease, and all of the participants were men.


The researchers also wondered whether giving more recovery time would change the magnitude of recovery they observed.


Nonetheless, according to the lead author, the study gives worthwhile insight into how we handle fat digestion.

New method found to inhibit Parkinson’s disease

Women drinkers at higher risk of liver disease

Women are more vulnerable to the damaging effects of alcohol than men because they are generally smaller in stature and have less body water, a US scientist has claimed.
According to Dr Howard Monsour, chief of hepatology at Houston Methodist Hospital, women who are already predisposed by genetics to have liver disease should limit their alcohol consumption or stay away from alcohol altogether.
"There is a misconception that you have to be an alcoholic to develop serious liver disease. Not true. In fact, if you have a genetic disposition, drinking more than a moderate amount could be very damaging, especially for women," Monsour said.
About 20 to 30 per cent of the population has a genetic disposition to cirrhosis of the liver and Monsour said it is important for people to know if they have a family history of cirrhosis before making the decisions to drink large amounts of alcohol.
"One drink a day might be too much for a woman who has a genetic pre-disposition to cirrhosis of the liver," Monsour said.
"One drink for a woman has about twice the effect as it does for the same amount consumed by a man," he said. The liver stores energy and nutrients and produces proteins and enzymes necessary for optimum health. It protects the body from disease and eliminates toxins like alcohol. "When women drink the same amount less is dispersed and the concentration is higher. They also have a lower activity of a metabolising enzyme in the stomach called alcohol dehydrogenase (ADH)," Monsour said.
ADH helps convert alcohol to acetaldehyde, which eventually is metabolised to carbon dioxide and water. This causes a larger amount of the alcohol to reach the blood and eventually in susceptible persons can lead to cirrhosis of the liver, a disease that normally has no visible signs until liver damage is too extensive.
Monsour added that people who think drinking a beer is better than hard liquor are misguided. One beer is equal to one shot of whiskey or one, four-ounce glass of wine. The alcohol content is the same in all three drinks.
