Researchers find antibody that blocks infection by covid in cell

Researchers have identified a fully human monoclonal antibody that prevents the SARS-CoV-2 (COVID-19) virus from infecting cultured cells.

The discovery, detailed in the journal Nature Communications, is an initial step towards developing a fully human antibody to treat or prevent the respiratory disease COVID-19 caused by the novel coronavirus SARS-CoV-2.

The COVID-19 pandemic has spread rapidly across the globe infecting more than 3.3 million people worldwide and killing more than 235,000 people so far.

This research from Utrecht University, Erasmus Medical Center and Harbour BioMed (HBM) builds on the work they have done in the past on antibodies targeting the SARS-CoV that emerged in 2002/2003.

“Using this collection of SARS-CoV antibodies, we identified an antibody that also neutralizes infection of SARS-CoV-2 in cultured cells,” said study co-lead author Berend-Jan Bosch, Associate Professor at Utrecht University in the Netherlands.

“Such a neutralising antibody has the potential to alter the course of infection in the infected host, support virus clearance or protect an uninfected individual that is exposed to the virus,” Bosch added.

The researchers noted that the antibody binds to a domain that is conserved in both SARS-CoV and SARS-CoV-2, explaining its ability to neutralize both viruses.

This cross-neutralising feature of the antibody is very interesting and suggests it may have potential in mitigation of diseases caused by future-emerging related coronaviruses, Bosch said.

This discovery provides a strong foundation for additional research to characterise this antibody and begins development as a potential COVID-19 treatment.

“The antibody used in this work is ‘fully human,’ allowing development to proceed more rapidly and reducing the potential for immune-related side effects,” said study co-author Frank Grosveld.

Conventional therapeutic antibodies are first developed in other species and then must undergo additional work to ‘humanize’ them, according to the researchers.

The antibody was generated using Harbour BioMed’s H2L2 transgenic mouse technology.

“This is groundbreaking research. Much more work is needed to assess whether this antibody can protect or reduce the severity of disease in humans,” said Dr Jingsong Wang, Founder, Chairman and Chief Executive Officer, Harbour BioMed.

“We expect to advance the development of the antibody with partners. We believe our technology can contribute to addressing this most urgent public health need and we are pursuing several other research avenues,” Wang noted.

Air pollution's tiny particles may trigger heart attacks

Researchers have found that even a few hours' exposure to ambient ultrafine particles common in air pollution may potentially trigger a non-fatal heart attack.

Myocardial infarction is a major form of cardiovascular disease worldwide. Ultrafine particles (UFP) are 100 nanometres or smaller in size. In urban areas, automobile emissions are the primary source of UFP.

For the study, published in the journal Environmental Health Perspectives, researchers investigated the effects of UFP exposure and heart attacks using the number of particles and the particle length and surface area concentrations at hourly intervals of exposure.

"This study confirms something that has long been suspected--air pollution's tiny particles can play a role in serious heart disease. This is particularly true within the first few hours of exposure," said the study's first author Kai Chen, Assistant Professor at Yale University in the US.

UFP constitute a health risk due to their small size, large surface areas per unit of mass, and their ability to penetrate the cells and get into the blood system, the study said.

The researchers were interested in whether transient UFP exposure could trigger heart attacks and whether alternative metrics such as particle length and surface area concentrations could improve the investigation of UFP-related health effects.

The research team examined data from a registry of all non-fatal MI cases in Augsburg, Germany.

The study looked at more than 5,898 non-fatal heart attack patients between 2005 and 2015.

The individual heart attacks were compared against air pollution UFP data on the hour of the heart attack and adjusted for a range of additional factors, such as the day of the week, long-term time trend and socio-economic status.

"This represents an important step toward understanding the appropriate indicator of ultrafine particles exposure in determining the short-term health effects, as the effects of particle length and surface concentrations were stronger than the ones of particle number concentration and remained similar after adjustment for other air pollutants," said Chen.

"Our future analyses will examine the combined hourly exposures to both air pollution and extreme temperature. We will also identify vulnerable subpopulations regarding pre-existing diseases and medication intake," Chen added.

Newly discovered antibiotics kill bacteria differently

Antibiotic resistance is a growing public health problem across the globe, with many diseases becoming harder to treat. Now, a newly discovered antibiotic group shows promise in the fight against superbugs as it has a unique way of killing bacteria.

A team of scientists at McMaster University has found a new group of antibiotics that can fight infections in a new and unique way. These antibiotics fight infections in a way researchers have never seen before, according to the findings of the study described in the journal Nature.
Holy grail' of antibiotics

The newly found group of antibiotics, consisting of corbomycin and complestatin, can kill bacteria by blocking the function of the bacterial cell wall. These drugs come from a family of antibiotics known as glycopeptides, which are produced by soil bacteria.

The two antibiotics attack peptidoglycan, the main component of the bacterial cell wall that is vital to the growth and survival of almost all bacteria. They inhibit the action of autolysins, which are important for cell division and growth.

Other antibiotics, such as penicillin, work by preventing the bacteria from building its wall, which is the source of its strength. In killing the bacteria, removing its wall will make it vulnerable and easier to kill.

These new antibiotics work by doing the opposite. Instead of preventing building the wall, it halts the wall ll from being broken down. As a result, blocking the breakdown of the wall would make it impossible for them to divide and expand – just like being trapped in prison.


Unique bacteria killer
The two new antibiotics are known as glycopeptides. The team studied the genes of the group to see if they lack resistance mechanisms. The team believes that if the genes that made these drugs different, perhaps the way they kill will also be different.

In collaboration with scientists from the Université de Montréal, including Yves Brun, they found that the drugs act on the bacterial cell wall to prevent it from dividing and proliferating. 

"Knowing the detailed structure at the atomic level of this connection between the surface layer and the surface of the cell offers enormous potential to then develop molecules that can target this attachment and make the cell more sensitive to antibacterials," Yves Brun, study co-author, said.

"Combined with the discovery of the new mode of action of two antibiotics, this development opens up prospects for weakening the action of bacteria and making them more vulnerable," he added.

The researchers believe the group of drugs is a promising clinical candidate in the hopes of stemming bacteria from becoming resistant to antibiotics.


Fight against antibiotic resistance

Antibiotic resistance is one of the greatest threats to global health, according to the World Health Organization (WHO). Though it happens naturally, the misuse of antibiotics is hastening the process, making it easy to treat infections in the past harder to curb now.

Further, antibiotic resistance increase hospital stays and medical costs. For instance, diseases in the past that were responsive to certain antibiotics may become resistant and difficult to stem, such as tuberculosis, pneumonia, gonorrhea, and other infections. Now, as the diseases become stronger and more resilient, outbreaks may become inevitable, unless new drugs are discovered.

In the United States alone, at least 2.8 million people become infected with antibiotic-resistant bacteria each year, while more than 35,000 people die.

Postmenopausal women may suffer from poor sleep quality, sleep disorders and shorter sleep duration

 Sleep is extremely important. It is a break from the long day at work, the end of the day, and gives us a new day to look forward to when we wake up. Biologically, it is restorative and helps the body recover from diseases, disorders, tiredness, fatigue, etc. A good night's sleep is especially important when you have had a long day, are extremely tired, are not keeping well, or are going through transition phases physically.

One such transition phase is that of menopause. Menopause is when the production of hormones responsible for reproduction comes to a halt in a woman's body. The body experiences various changes through the phase of menopause. A recent study has found that sleep complications are also common during the menopausal phase.

The study was published online in Menopause – the journal of The North American Menopause Society. The study compared sleep quality, duration and disorders between postmenopausal and premenopausal women, and found that sleep problems increase postmenopause.

Statistically, also, sleep disorders are more common during menopause and affect 40 to 60 per cent women.

Poor sleep quality, disorders, and short duration sleep can have adverse health effects. They lead to poor quality of life, high risk of cardiovascular diseases, diabetes, depression, and anxiety. Since certain sleep disorders like sleep apnea are also age-related and are more common in people of older age, women of postmenopausal age are at an elevated risk of such disorders as well.

The study found that postmenopausal women required more time to fall asleep, an excess of thirty minutes when compared with premenopausal women. They were also found to be more likely to suffer from insomnia and obstructive sleep apnoea.
