Researchers Have Identified Why Our Bodies Reject Transplanted Organs

Researchers have identified a previously hidden link between our immune system and the activation of cells that lead to organ rejection.

The discovery opens the way for scientists to develop new forms of treatment that could prevent immune responses from attacking life-saving tissue transplants without leaving the body so open to infection or cancer.

A type of receptor on bone marrow cells called signal regulatory protein alpha  (SIRPα) has been identified by scientists, as the body's watchdog responsible for dispatching the lymphocytes that target and destroy foreign cells.

In simple terms, whenever we take cells from another person's body and put them into our own, white blood cells see them as foreign and attempt to break them apart.

Not only is this bad news for the transplanted tissues, the swelling and fever that comes with the immune response isn't exactly a picnic either.

The mechanisms behind the white blood cell assault are fairly well understood – molecules on the outside of the cells belonging to what's called the major histocompatibility complex (MHC) identify them as different.

A type of white blood cell called a T lymphocyte has receptors on its surface capable of recognising unknown MHC proteins and responds by attempting to break up the foreign material they're attached to.

Lymphocytes aren't born knowing what's foreign and what's not; they need to be taught. Which is the job of another part of the immune system called a dendritic cell.

Dendritic cells chew up foreign proteins and weave them into their own MHC before displaying them on their surface like a microscopic 'wanted' poster. They then migrate into the body's lymph nodes where they interact with the gun-slinging T lymphocytes.

Exactly how dendritic cells identify foreign materials, however, has been something of a mystery.

To learn more, the researchers studied the transplanted tissues in mice engineered to lack certain white blood cells such as their T lymphocytes.

They found that differences between the mice donor's and recipient's SIRPα gene correlated with the recipient's immune responses.

SIRPα isn't an unknown protein, already understood to bind to another protein called CD47 that triggers a range of immune responses in different white blood cells.

Joining the dots, the researchers believe CD47 on monocytes – the white blood cells that grow into dendritic cells – interact with SIRPα receptors on foreign tissues, setting off the entire ID check process.

"Once these cells are activated, then they turn around and activate the rest of the immune system, and that leads to the full-blown rejection of the organ," lead researcher.

"What we would like to do is sequence the SIRP-alpha gene in many humans who are donors and recipients of organ or bone marrow, and then ask whether a mismatch affects the outcome after transplantation."

A better match between the donor's and recipient's SIRPα genes could help reduce the risk of the immune response being sparked in the first place.

Even when organs are chosen to have closely matching MHC proteins, subtle differences can still produce immune responses. For example, one in 10 hearts and nearly one in 20 kidneys will show signs of being rejected by the body within the first year in spite of being declared compatible.

To avoid rejection, organ recipients need to be on treatments that suppress the immune system, not only making them prone to infection and cancer but often raising the risk of heart attacks and strokes by increasing blood pressure.

New treatments focusing on SIRPα might help reduce the doses or types of medications, and possibly help prolong the organ's life.

Hepatitis- types and causes

In the simple terms, hepatitis means injury to your liver due to inflammation. There are five main types of hepatitis and the type that you are suffering from is determined by a laboratory test. All the five main types of hepatitis are caused by viruses. Out of the five, Hepatitis C and B are quite big. Globally, around 250 million people are affected by hepatitis C and 300 million people are estimated to be hepatitis B carriers.

Fact sheet on hepatitis
  1. Hepatitis A & E is caused by consuming contaminated water.
  2. Hepatitis B, C and D is transmitted through infected blood to blood contact.
  3. Hepatitis C is usually spread via direct contact with the blood of an infected person.
  4. The initial symptoms of hepatitis are the same as those of flu.
  5. Hepatitis can heal on its own causing no significant liver damage; however, if it progresses, it can cause extensive liver scarring and extensive damage of your liver.
Most liver damage is caused by 3 hepatitis viruses, and these are hepatitis A, B and C.
How do you safeguard yourself against infection from these three dangerous hepatitis viruses?

The most common cause of concern is that a lot of us who are infected with hepatitis, especially hepatitis B don’t know that we are. The biggest bulwark against hepatitis is periodic testing and vaccination. Vaccination is available for most hepatitis viruses including A and B, which tend to be extra virulent.
So, do take care and go for periodic testing. We are reiterating this because if you are infected with hepatitis B and are not treated you can develop serious problems of the liver, including liver cancer. In fact, nearly 1 in 4 people living with hepatitis B develop serious liver problems including liver cancer. Hepatitis B related liver cancer is a leading cause of fatality and getting tested for hepatitis can help you access life-saving treatments that can prevent serious liver damage.
Other ways to prevent hepatitis are
Hepatitis A
  1. Wash your hands with soap after using the toilet
  2. Only consume freshly-cooked food
  3. Only drink bottled or boiled water
  4. Only eat fruits that can be peeled. Don’t eat fruit that has been cut and kept
  5. Avoid eating raw vegetables
  6. Vaccinate yourself against hepatitis A if you travel a lot.
Hepatitis B
  1. Practice safe sex by using condoms
  2. Only use new syringes for injections
  3. Do not share toothbrushes, razors, or manicure instruments with others
  4. Vaccinate yourself against hepatitis B
  5. Take care while getting tattooed. Only use brand new tattoo needles. Also, ask for new needles during acupuncture treatments.
Hepatitis C
  1. Avoid sharing toothbrushes, razors, and manicure equipment
  2. Cover open wounds if you are infected
  3. Don’t share needles. Always ask for fresh needles.
  4. Use only well-sterilized equipment for acupuncture, piercing, and tattooing etc.
  5. Cut down on alcohol
  6. Do not share drug needles
You must consult your physician in case you've any doubt or if you're having any of the above symptoms.

6 Types of Hypertrophic Scar Treatment

One of the most unpleasant scars to deal with is a hypertrophic scar, which can cause disfigurement that can lead to social detachment, lowered self esteem and job discrimination. This type of scar is extremely visible and tends to be raised above the surrounding skin.

What Is A Hypertrophic Scar?
This type of scar is raised, thick, red and is usually caused by injury or even surgery. They are usually the body's response to inflammation, surgery, trauma or burns. This type of scarring is most common among young people, particularly those with a darker skin tone. This type of scar forms when something goes wrong during the initial phase of healing and causes an overabundance of collagen fibers in the affected area. Due to the accumulation of collagen, the scar looks raised in appearance instead of flat like other scars.

Most often, they tend to form on the breastbone, shoulders and ears but can also form on other parts of the body. Other characteristics of this type of scarring include itchiness and pain, even after the wound has healed.

Hypertrophic Scars and Keloid Scars: Are They Similar?
Similar to hypertrophic scars, keloid scars can be caused by injury, surgery, injections, body piercing and also acne. They are both similar in size and shape, though keloid scars will grow beyond the boundary of the wound. Both scars have high amounts of collagen in comparison to the average scar tissue. In contrast, hypertrophic scars can heal by themselves within 12 to 18 months, unlike keloid scars, although the scar does not disappear completely.

What Are Hypertrophic Scar Treatment Options?
Even though hypertrophic scars can heal with no assistance within several months, certain treatment options can help to accelerate the process and minimize the appearance of the scars. Below are treatment options you could try:

1. Needling Hypertrophic Scar Treatment:  This affordable and cost-effective treatment can be done at home with specialized scarring needles. The treatment requires that you 'needle the scar' often in order to stimulate the formation of the collagen fibers. Seek medical care at first and enquire about the possible side effects.

2. Collagen Injections: Such an injection can be used on scars that sink into the skin rather than grow out of it. It requires injecting the collagen into the sunken scar which will help it to rise above skin level.

3. Steroid Injections: This scar treatment is used by many people as it helps thin the skin tissues. The steroid is injected directly into the scar every month until it has flattened out. Although effective it can pose certain risks.
4. Silicone Sheets Hypertrophic Scar Treatment: This is considered to be one of the best treatments as it helps to fade most of the traces of the scarring. It is usually performed by a dermatologist but can also be done as a home treatment. Here, silicone gel sheets are applied to the skin to moisturize the scar and even out the blotchy skin tone around it. They are mineral-based gel membranes, transparent and very smooth, making them easy and comfortable to wear. The sheets cover the scar by applying pressure, though air and moisture can still penetrate the sheets. This treatment is very low-risk and does not cause further damage to the skin tissues.

5. Radiotherapy: In severe cases, radiotherapy treatment is performed whereby the scar is cut open and closed up or exposed to radiotherapy. It is very risky, however, as opening a scar can have long-term side effects.

6. Laser Therapy:  This form of therapy effectively removes all kinds of scars including hypertrophic acne scars. It works by vaporizing the scar tissues one level at a time. Treatment starts with the top layers of the skin, till the lower skin layers are exposed. After a couple of sessions, the scar should disappear - though this would depend entirely on the severity of the scar.

Diet For Crohn's Disease

A study has shown that a high fat diet could lead to specific changes in gut bacteria that may help fight Crohn's disease.

This is because “good” fats help tackle harmful inflammation.

Since Crohn’s is a type of inflammatory bowel disease, the research might prove a major discovery for sufferers.

There are currently many people who are suffering from the condition where the underlying cause remains unknown.

Symptoms include intestinal swelling, cramping, and diarrhoea.

However, the researchers discovered that a diet of plant-derived "good" fats could reduce bacterial diversity.

Such foods include the coconut oil and cocoa butter that was used in the study, as well as avocados, nuts, salmon and olive oil.

In the study, mice fed fatty diets experienced up to 30 per cent fewer kinds of gut bacteria as their counterparts who were fed a normal diet - this resulted in a very different gut microbial composition.

Even those fed low concentrations of coconut oil or cocoa butter had less severe small intestine inflammation.

The researchers said: “The finding is remarkable because it means that a Crohn's patient could also have a beneficial effect on their gut bacteria and inflammation by only switching the type of fat in their diet.

“Patients would only need to replace a 'bad' fat with a 'good' fat, and eat normal amounts."

Researchers believe the results could lead to new anti-inflammatory probiotics.

This is because they may be able to now identify bacteria to use which can treat patients suffering from inflammatory bowel syndromes.

The study is one of the first to identify specific changes in gut bacteria - or microbiome - associated with Crohn's disease.

Additionally, it is the first to show how high fat diets can alter gut bacteria to combat inflammation.

A researcher said: “Ongoing studies are now helping us to understand which component of the 'good' and 'bad' fats make the difference in the gut microbes and make mice healthier.

"Ultimately, we aim to identify the 'good' fat-loving microbes for testing as probiotics."

SOME COMMON CAUSES OF NECK PAIN

Neck pain is a common  problem which many of us face these days. It usually occurs due to strained neck muscles, often caused bad posture, though there might be other causes as well. Rarely, neck pain can be a symptom of a more serious problem as the condition is relatively mild in nature. The symptoms of a neck pain are very obvious which refers to any kind of discomfort in moving your neck on a daily basis. Along with this you might also have headache, stiffness of the head and muscle tightness.
Some of the most common causative factors of neck pain are listed below:
  1. Muscle strains: A muscle strain generally happens when you exert too much pressure on your neck continuously for long periods. It also happens when you have your neck fixed in the same position for long hours of the day. Muscle strains in the neck may occur when you keep your neck bent in the wrong way while doing activities like reading a book, looking at your mobile phone or working over your laptop for extended hours.
  2. Worn-out joints: Just like all the other bodily joints, the neck joint also tends to wither and degrade with age. This phenomena is known as the wear and tear of the joints. The bones of the neck joint tend to wither with time, thereby making your joint frail and exposing it to various pains and medical conditions.
  3. Nerve compression: Nerve compression is a medical symptom whereby, there is excessive pressure put on one particular nerve in the body. Herniated discs or bone spurs in the vertebrae of your neck can press on the nerves branching out from the spinal cord, hence causing a long lasting neck pain. Damage caused from a nerve compression may be minor or severe depending on the pressure applied. It may cause temporary or long lasting problems depending on the degree of damage caused. The earlier you get a diagnosis and treatment for nerve compression, more quickly you will find relief.
  4. Whiplash injuries: Neck related injuries are common phases of pain and discomfort caused due to an external damage to the neck as a result of a tragic sports related or vehicular accident. A neck injury is also known as a whiplash and it refers to an injury due to forceful, rapid back-and-forth movement of the neck, like the cracking of a whip. Whiplash mostly occurs during accidents, physical abuse or other trauma.                   If you've pain due to any of these conditions, kindly consult your physician or orthopedic to get relief.

Blocked Spleen can cause chest pain !

Spleen lies behind our left rib-cage and helps protect your body from infection. It’s rare, but blood flow to your spleen can get blocked because of a blood clot, infection, or disease. If that happens, the tissue there can start to die. This is called a splenic infarction. Some people have no symptoms, but others have chest pain, often on their left side. It can get better with medication but can become serious if it’s not treated. If you've any such symptoms, kindly get in touch with your physician to rule out all possibilities.
