Protein markers in breast milk may help for early detection of cancer

Protein markers in breast milk can help detect breast cancer at an early stage and even predict if a woman is at risk of the deadly disease, a new study has found. Objective detection of breast cancer in young women in the early stages is challenging as mammography and imaging techniques are less effective because young women have dense breast tissue and there is a possible relationship between pregnancy and BC risk, researchers said.

One of the potential options for detection of BC is biochemical monitoring of protein markers in different types of bodily fluids such as serum, nipple aspirate fluid and ductal lavage fluid, tear, urine, saliva and breast milk. Researchers, including those from University of Massachusetts Amherst in the US, investigated biochemical signatures of breast cancer that are detectable in breast-milk.

 They compared breast-milk samples from women with breast cancer, women without breast cancer and women who were later diagnosed with breast cancer.

 The team then identified alterations in protein expression in breast-milk when a woman has – or will soon develop – breast cancer which might be due to cancer risk or development. Breast milk provides access to breast tissue in the form of exfoliated epithelial cells that are the origins of the most types of breast cancers, researchers said. Analysis of breast milk is a non-invasive method that could be used for cancer diagnosis.


After further validation, the method could offer a new, non-invasive approach to breast cancer screening for women in their childbearing years.

Anti-ageing serum can do more harm than good for elderly people

A team of researchers has found that by removing senescent cells, it may bring us one step closer to the "fountain of youth". However, experts warned that an anti-ageing serum could be a few years off yet as the drugs may be unsafe for elderly people.
 
A drug has previously been found to help elderly mice regrow their hair, run faster and live for longer. It works by removing cells in skin tissue that naturally accumulate as the rodents grow older.

But the senescent cells - which are unable to reproduce themselves and prevent tissue growth - are also found in humans. The study was published in the journal Trends in Molecule Medicine.

"This strategy may bring us one step closer to the " fountain of youth," but it's important to be cautious and not hype," said researcher of aging Peter de Keizer of the Erasmus University Medical Center in the Netherlands.

They noticed that these permanently arrested cells accumulate in mature tissue and that some of them secrete factors that are harmful to tissue function and impair their neighboring cells.

To explain what causes this noise in the system, de Keizer proposes a "senescence-stem lock model" in which the chronic secretion of pro-inflammatory factors by these senescent cells keeps neighboring cells in a permanent stem-like state and thereby prevents proper tissue renewal.

"A perfect anti-senescence therapy would not only clear senescent cells, but also kick-start tissue rejuvenation by stimulating differentiation of nearby stem cells. This may be complementary with, for instance, the exciting approaches recently made in the field of transient expression of stem cell factors," de Keizer stated.
There's still much basic research to be done before humans visit their local rejuvenation clinic for their annual shot of anti-aging serum.

Senescent cells do have a temporary role in wound healing, so you don't want to eliminate them when you are injured or at the wrong point in time.

"I would also advise caution for claiming too much, too soon about the benefits of the fast-growing list of therapeutic compounds that are being discovered. That being said, these are clearly very exciting times, and I am confident we will find applicable anti-senescence treatments that can counteract age-related pathologies," de Keizer explained.

Researchers will also need to think about when such treatments should be administered (such as before or after the onset of certain conditions) and who would benefit the most.

The potentially high cost of an anti-aging therapy, as well as off-target toxicity, could also be limiting factors for widespread market use as it is translated.

De Keizer, who plans to co-found a start-up based on the discovery of anti-senescence compounds from his lab, is hopeful that cell-penetrating peptides that can block specific activities of these retired cells could be the path forward over broad-range inhibitors.

High vitamin B level during pregnancy cuts eczema risk in kids

Eating foods rich in a type of vitamin B - such as fish, meat, chicken and mushrooms - during pregnancy may lower the risk of your child developing eczema, a condition that causes the skin to become dry and itchy, a new study has found.

The study from the University of Southampton in the UK is the first to link maternal serum levels of nicotinamide, a naturally occurring vitamin, and related metabolites to the risk of atopic eczema in the child.



The researchers believe the findings support the concept that eczema partly originates as a baby develops in the womb and could show ways to reduce the risk of the skin condition.

"Nicotinamide cream has been used in the treatment of eczema but the link between the mother's levels of nicotinamide during pregnancy and the offspring's risk of atopic eczema has not been previously studied," said Sarah El-Heis, from the University of Southampton.

"The findings point to potentially modifiable influences on this common and distressing condition," said El-Heis.

Nicotinamide is a form of vitamin B3. Its level is maintained through intake of foods such as fish, meat, chicken, mushrooms, nuts and coffee as well as tryptophan, an amino acid found in most proteins.

Nicotinamide and related nutrients are important for the body's immune responses and energy metabolism.

The research assessed the amount of nicotinamide and related tryptophan metabolites during pregnancy in 497 women that took part in the Southampton Women's Survey. The rates of eczema in their children at ages 6 and 12 months was studied.

Results showed that offspring of mothers with higher levels of nicotinamide had a 30 per cent lower chance of developing atopic eczema at 12 months.

There was an even stronger association with higher levels of anthranilic acid, a tryptophan metabolite.

Nicotinamide can improve the overall structure, moisture and elasticity of skin and therefore could potentially alter the disease processes associated with eczema, researchers said.

The study showed a gradual association between higher maternal nicotinamide and anthranilic acid levels and a lower risk of atopic eczema, suggesting that the development of eczema is not simply prevented by the presence of these nutrients.

"More research is needed to investigate this interesting association, but the findings are further evidence of the potential benefits of eating a healthy balanced diet during pregnancy," said Keith Godfrey, Director of the Southampton Biomedical Research Centre in Nutrition.

The study was published in Clinical and Experimental Allergy.

Walnuts may prevent diabetes and heart disease

Eating walnuts daily can ward off diabetes and heart disease in at-risk individuals, a new study has found.
Researchers  found that daily intake of 56 g of walnuts improves endothelial function in overweight adults with visceral adiposity.
The study included a sample of 46 adults aged 30-75.
Participants had a Body Mass Index larger than 25, and a waist circumference exceeding 40 inches for men and 35 inches for women.
They were also required to be non-smokers, and all exhibited one or more additional risk factors for metabolic syndrome, a precursor of diabetes and cardiovascular disease.
The group was randomly assigned to two 8-week sequences of either a walnut-enriched ad libitum diet or an ad libitum diet without walnuts.
Those chosen for the walnut diet were instructed to consume 56 g of shelled, un-roasted English walnuts per day as a snack or with a meal.
“We know that improving diets tends to be hard, but adding a single food is easy,” explained Dr and lead author of the research team.
“Our theory is that if a highly nutritious, satiating food like walnuts is added to the diet, there are dual benefits: the benefits of that nutrient rich addition and removal of the less nutritious foods,” Dr. said.
The research found that daily intake of 56 g of walnuts improves endothelial function in overweight adults with visceral adiposity.
“The primary outcome measure was the change in flow-mediated vasodilatation (FMD) of the brachial artery,” researchers said.
“Secondary measures included serum lipid panel, fasting glucose and insulin, Homeostasis Model Assessment-Insulin Resistance values, blood pressure, and anthropometric measures.
“FMD improved significantly from baseline when subjects consumed a walnut-enriched diet as compared with the control diet. Beneficial trends in systolic blood pressure reduction were seen, and maintenance of the baseline anthropometric values was also observed. Other measures were unaltered,” they said.
