Influence of genetics on cannabis dependence in adolescent females

Researchers have identified that a variant in a particular gene, that is found in about one-third of people, increases vulnerability to THC in females. It also has a large-scale impact on brain regions and pathways responsible for processing reward.

The brain's endocannabinoid system regulates the activity of cannabinoids that are normally produced by the body to influence brain development and regulate mood as well as those from external sources, such as the psychoactive ingredient THC, also known as D9-tetrahydrocannabinol, which is found in cannabis.

An enzyme called fatty acid amide hydrolase (FAAH) breaks down a cannabinoid called anandamide that is naturally found in the brain and is most closely related to THC, helping to remove it from circulation.

In the study which was published in 'Science Advances,' the researchers examined mice harbouring a human gene variant that causes FAAH to degrade more easily, increasing overall anandamide levels in the brain.

From the preclinical research by Weill Cornell Medicine investigators, they discovered that the variant resulted in an overactive reward circuit in female--but not male adolescent mice--that resulted in a higher preference for THC in females.

Dr. Caitlin Burgdorf, recent doctoral graduate from the Weill Cornell Graduate School of Medical Sciences, and the lead author said "Our study shows that a variant in the FAAH gene, which is found in about one-third of people, increases vulnerability to THC in females and has large-scale impact on brain regions and pathways responsible for processing reward."

"Our findings suggest that genetics can be a contributing factor for increased susceptibility to cannabis dependence in select populations," Caitlin added.

The result showcased that female mouse with the FAAH variant showed an increased preference for the environment in which they'd been exposed to THC over a neutral environment when they were exposed to the substance during adolescence, and the effect persisted into adulthood.

Making Art Is Good For Your Health. Mke it a habit

Whether it's woodworking, sewing or sculpting teeny cupcakes out of bright purple clay, making art is good for your mind and body.

The act of creation can reduce stress and anxiety and improve your mood, says Girija Kaimal, a professor at Drexel University and a leading researcher in art therapy. And flexing our creative side can give us a stronger sense of agency — the ability to solve problems by imagining possible solutions. 


Making art can also feel pretty awesome, she says. Engaging in any act of visual expression activates the reward pathway in your brain, "which is perceived as a pleasurable experience."

Artists say they often feel those positive vibes when they work. Trinidad Escobar, a Filipino American graphic novelist and poet based in Oakland, Calif., says that drawing "feels like a superpower." And Liana Finck, a cartoonist for The New Yorker, says sharing her art with an audience is "the most cathartic thing in the world."

But you don't have to be a full-time artist to get in on those health benefits, says Kaimal. All you need to do is just make art. And the more you do it, the better. Here are six things you need to know to kick-start your creative art habit.


1. You don't need to be an artist with a capital "A"
We tend to think that only people who are very skilled at art can call themselves artists, but really, anyone can be an artist, says Kaimal.

"Everyone is capable of creative expression," she says. In fact, her own research has shown that there are no differences in health outcomes between those who identify as experienced artists and those who don't. So that means that no matter your skill level, you'll be able to feel all the good things that come with making art. 


Christianne Strang, a professor of neuroscience at the University of Alabama at Birmingham, promises that if you stick with it, you'll enjoy it. In her 30-year career as an art therapist, she has found that "99% of the time, people find that if they give up the idea that they're not good enough, if they give up the judgment, making art actually feels good."


2. Figure out your mode of creative expression
Start with what you enjoy — maybe something you've done before, maybe something you loved as a kid. But keep an open mind.

"Anything that engages your creative mind — the ability to make connections between unrelated things and imagine new ways to communicate — is good for you," says Kaimal.

You can do that through myriad activities: finger painting, cooking, baking, collaging, oil painting, weaving, knitting, crocheting, writing screenplays, scrapbooking — the sky's the limit. And don't feel like you have to stick to one thing, say the art therapists. Mix it up — do whatever you're in the mood for. 


3. Focus on the making, and let go of expectations

Once you have your art materials, here comes the fun part. Mess around! Let yourself get lost in the process and just play. Because it's the physical act of making art, says Kaimal, that induces those feelings of stress relief and positive energy — not what you make or how you make it.

Kaimal says you don't even need to complete a project or like what you're making to feel those health benefits.

In fact, says Strang, that can be a positive thing. In her free time, she likes doodling, watercoloring and glass fusing. "Sometimes, when I don't like what I'm making, it frees me up to play and explore new ideas, because I've let go of expectations," she says.
 

4. Think about making art like any healthy habit, such as eating well or exercising

Just as you make time to work, exercise and hang out with family and friends, you should make time for your artistic endeavors, says Strang. "Creativity in and of itself is important for remaining healthy — remaining connected to yourself and remaining connected to the world," she says.

While there are no hard and fast rules for how much time you should devote to engaging in art, Strang has a suggestion: "as much as you can get away with."

Try to designate a special time in your week to devote to your practice, says Kaimal. She likes to set aside time on the weekends to work on her own art projects: mixed-media pieces inspired by nature. She calls it her "TGIS." "Thank God it's Saturday," she says. "It's my few hours of time to develop something I'm working on and really take that time for myself."

You don't even need a few hours. Escobar, the Oakland-based artist, teaches community art classes to people of color and members of the LGBT community. She says that doing just 10 minutes of art each day can do wonders.

Whatever you do, don't wait for creative inspiration to strike. "That's a myth that you'll create because you're in this altered state of mind and feeling free and loose," she says.

In addition to carving out time, carve out a physical space in your home for art, says Kaimal. She suggests setting up a corner table for your sketchbook and art supplies. If space is an issue, put it all in a basket or a plastic container that's easily accessible.

Your creative corner is "kind of like a reminder," she says. "It reinforces your commitment to invest in yourself and your well-being." 


5. Ride those waves of emotion
The more you're able to make art a regular habit, the more you're likely to get this great reward: this wonderful thing that happens when you're in the zone. Kaimal says that scientists have a word for it. It's called "flow."

"It's that sense of losing yourself, losing all awareness. You're so in the moment and fully present that you forget all sense of time and space," she says.

Escobar says she feels that flow when she's filling in lines with color, inking — which in the world of cartooning means tracing over pencil marks with ink — and when she's drawing aesthetically pleasing backgrounds, such as jungle or garden scenes.

"It feels meditative," she says, like things are "going well, smoothly, like the process is nourishing me."


6. Get yourself in the mood

Some days, you won't feel like making art. And that's OK, says Finck. As the author of several graphic novels, most recently Excuse Me: Cartoons, Complaints, and Notes to Self, she's used to juggling multiple projects.

"I get a lot of writer's block," she says. "It feels like stage fright when I sit down with a blank page."

To trick herself into drawing, Finck tells herself to think "that I'm just doodling," she says. "Then it flows freely."

If the block persists, she checks in with herself. What's stopping her from drawing? Is she anxious? Depressed? Worried?

"If you're too anxious to draw, address the anxiety. Go take a walk. Stand up from the couch. Go for a run. See a person," says Finck.

You can also try a breathing exercise, says Escobar. "Inhale for four seconds and hold for a second. Repeat that for about three minutes."

She does this with students in her art classes to help them reset and refocus after a long day at work — and it works, she says, like a charm.

Alzheimer's: Head device reduces memory loss in 7 out of 8 people

An open-label clinical trial working with eight people with Alzheimer's disease concluded that a new wearable device that emits electromagnetic impulses was able to significantly improve memory loss in seven of these participants within 2 months. 

Alzheimer's disease affects millions of people in the United States and worldwide, but as yet there is no cure for this progressive neurodegenerative condition.

The condition's main physiological characteristic is the aggregation of beta-amyloid and tau protein in the brain, which disrupt normal communication pathways between brain cells.

Scientists have been aware of this aspect of Alzheimer's for years but have so far been unable to prevent the aggregates from forming, or to dissolve them once formed — not in humans, at least.

But now, researchers affiliated with NeuroEM Therapeutics — a medical device company based in Phoenix, AZ — have developed a wearable device that, according to their recent open-label clinical trial, can significantly reduce memory loss in Alzheimer's by disaggregating toxic protein formed in the brain.

The device has the shape of a cap, and it emits electromagnetic waves in a frequency that, as preclinical studies in mice have shown, can help reverse memory loss. The research team that conducted the clinical trial reports the groundbreaking results in a study paper .

"Despite significant efforts for nearly 20 years, stopping or reversing memory impairment in people with Alzheimer's disease has eluded researchers," notes one of the researchers, Dr. Amanda Smith, who is the director of clinical research at the University of South Florida Health's Byrd Alzheimer's Institute in Tampa.

    "These results provide preliminary evidence that [transcranial electromagnetic treatment] administration we assessed in this small [Alzheimer's disease] study may have the capacity to enhance cognitive performance in patients with mild to moderate disease."

'Highly significant improvement' at 2 months

For the trial, the researchers worked with eight participants with mild to moderate Alzheimer's disease and their caretakers, who received instructions on how to use the therapeutic device at home. The participants received the treatment twice a day for 2 months, and each session lasted just 1 hour.

By the end of the 2 months, none of the participants had experienced any side effects. Brain scans conducted by the researchers at the end of the study showed that the eight individuals had not developed tumors or brain bleeding as a result of having used the device.

To assess whether the treatment had helped the participants, the investigators used The Alzheimer's Disease Assessment Scale-Cognitive Subscale test (ADAS-cog), the most widely recognized method of assessing cognitive function.

The team found that seven of the eight participants saw an increase of over 4 points in cognitive performance on the ADAS-cog scale after 2 months. This, the researchers explain, is as if the participants' cognitive function had "rejuvenated" by a year.

"We were particularly surprised that this highly significant improvement in the ADAS-cog was maintained even 2 weeks after treatment was completed," says Dr. Gary Arendash, CEO of NeuroEM Therapeutics. "The most likely explanation for continued benefit after cessation of treatment is that the Alzheimer's Disease process itself was being affected," he adds.

The researchers also collected blood and cerebrospinal fluid samples from the participants both at baseline and at the end of the clinical trial.

In analyzing them, they found that the intervention appeared to have led to the disaggregation of beta-amyloid plaques and tau tangles, which are associated with progressive cognitive function impairment in Alzheimer's.

Furthermore, MRI scans also suggested that after the 2-month treatment period, the participants had better communication between brain cells present in the cingulate cortex, which plays a key role in cognitive function — including decision making.


Further steps

The researchers behind the innovative device also note that they received the best feedback they could have received: all of the participants decided to hang onto the devices they got as part of the clinical trial.

"Perhaps the best indication that the 2 months of treatment was having a clinically important effect on the [Alzheimer's disease] patients in this study is that none of the patients wanted to return their head device to the University of South Florida/Byrd Alzheimer's Institute after the study was completed," says Dr. Arendash.

The team is particularly excited about the device's effect on toxic brain plaques. They say that drugs tested in clinical trials have, so far, been much less successful in disaggregating these disruptive buildups.

So, the investigators do not plan on stopping at this small clinical trial. Going forward, they have offered the current trial's participants' the opportunity to take part in a much larger clinical trial, which the research team is now organizing.


All the former participants have accepted this offer. The new trial is due to last around 17 months, on average, and will include about 150 participants with a diagnosis of mild to moderate Alzheimer's disease.

The medical device company hope that they will be able to gain Food and Drug Administration (FDA) approval for their technology over the next couple of years and make the device available to the public by 2021.

Fatty liver disease can sometimes lead to liver cancer

In a new study, researchers have discovered important differences in the biological pathways that lead to cancer for alcoholic fatty liver disease compared to non-alcoholic fatty liver disease. The findings of the study, could benefit more than 3 million people diagnosed with the fatty liver disease each year. The build-up of fat in the liver known as fatty liver disease sometimes leads to hard-to-treat liver cancer.

Scientists have not been able to understand why the cancer risk is higher for fatty liver disease caused by excessive alcohol consumption than for non-alcoholic fatty liver disease, which is associated with obesity and type 2 diabetes.

"We want to find out the key factors and pathways that lead to liver cancer in fatty liver patients so we can slow or even prevent the liver cancer. It is estimated that this type of liver cancer is responsible for 250,000 to 1 million deaths each year," said the author who conducted the study.

In the study, the researchers examined expression levels of proteins in liver biopsies from a group of patients with alcoholic fatty liver disease and a group with non-alcoholic fatty liver disease.

The proteins studied are involved in epigenetic regulation, which controls gene expression, and inflammation, which plays an important role in the disease.

The researchers observed that the two groups of patients showed significant differences in the expression of proteins involved in epigenetic regulators and inflammation.

Importantly, these differences matched the ratio of liver cancer development seen for the two groups."If through additional studies, we can prove that the molecules and pathways we identified modulate liver cancer development in the alcoholic fatty liver disease or non-alcoholic fatty liver disease patients, it may help identify new targets for preventing or reducing the risk of liver cancer that arises from other liver diseases," he said.

The researchers plan to perform additional experiments in animal models of fatty liver disease to learn more about how epigenetic regulation and inflammation pathways lead to liver cancer.

Eating walnuts may help fight breast cancer

Consumption of walnuts may help suppress growth and survival of breast cancer, a study claims.
The study, found that consumption of two ounces of walnuts a day for about two weeks significantly changed gene expression in confirmed breast cancers.

“Consumption of walnuts has slowed breast cancer growth and reduced the risk of mammary cancer in mice,” said a researcher.

“Building on this research, our team hypothesised that walnut consumption would alter gene expression in pathologically-confirmed breast cancers of women in a direction that would decrease breast cancer growth and survival,” the researcher said in a statement.

In this first clinical trial, women with breast lumps large enough for research and pathology biopsies were recruited and randomised to walnut consuming or control groups.

Immediately following biopsy collection, women in the walnut group began to consume two ounces of walnuts per day until follow-up surgery.

Pathological studies confirmed that lumps were breast cancer in all women who remained in the trial.
At surgery, about two weeks after biopsy, additional specimens were taken from the breast cancers.

Changes in gene expression in the surgical specimen compared to baseline were determined in each individual woman in walnut-consuming and control groups.

RNA sequencing expression profiling revealed that expression of 456 identified genes was significantly changed in the tumour due to walnut consumption.

The study showed activation of pathways that promote apoptosis or programmed cell death and cell adhesion and inhibition of pathways that promote cell proliferation and migration.

“These results support the hypothesis that, in humans, walnut consumption could suppress growth and survival of breast cancers,” he  said.

“Additional research through a larger-scale study would be needed to clinically confirm that walnut consumption actually does reduce the risk of breast cancer or breast cancer recurrence,” said the researcher.

A large protein found in spinach may aid in the development of new medications for millions around the world dealing with alcohol use disorders, chronic pain, and mood disorders, researchers said. The study, discovered two peptides which are naturally metabolic products of Rubisco — a large protein found in many plants like spinach, which will aid within the development of latest medications. “These disorders are presently not adequately managed,”according to the Prof.

The researchers are actively pursuing synthetic and computational strategies to improve these peptides to make them more effective. The rubiscolin peptides are also being investigated for their ability to regulate dietary intake and are even commercially available in anti-aging skin products.

Scientists Explain Connection Between Sugar and Cancer Growth

Most of us love sweets. Sugar is not just present in desserts, but also in all soft drinks, including colas. Having the soft drinks is considered as having empty calories, but it isn't just empty calories that scientists are worried about, but also about its effect on the body, especially, the cancer cells feed on sugar to thrive.

Unfortunately, even small amounts of those sneaky sugar molecules can be connected to cancer growth in our bodies. With an eye on prevention, scientists are discovering more about how and why sugar affects cancer cell growth and what we can do to distance ourselves even further from sugar’s unhealthy effects.

Sugar is a carbohydrate molecule, often associated with sweet treats, but it can be found in natural fruits as well. Our bodies use carbohydrates as fuel, and sugar can be a complex or simple carbohydrate molecule. Complex carbohydrates exist in starches like pasta and potatoes. They are basically a bunch of sugar molecules strung together. The difference is that whereas complex carbohydrate foods contain healthy fiber, vitamins and minerals, simple carbohydrates like white sugar and high fructose corn syrup have none of those nutritional benefits.

Do we need sugar to survive? Not exactly. As long as we have carbohydrates in our diet, we can avoid sugar. However, with the wide variety of foods containing naturally occurring sugars, like lactose and fructose for example, we would eliminate a lot of tasty and nutritional items by avoiding all dietary sugars.

Sugar is an acidic molecule

Acidity is a happy place for cancer and disease in the body, so no wonder an acidic molecule like sugar is connected to cancer growth. The more sugar we consume, the more acidic our bodies are. The more acidic our bodies are, the more likely disease will spread because it is being kept in its favorite climate. It’s like any existing cancer cells in your body get to go on an all-inclusive vacation at the beach in Fiji and multiply like bunnies whenever you have lots of sugar in your diet.

Dr. Otto Warburg first associated cancer with an acidic environment back in 1931, and he won a Nobel prize in Physiology or Medicine for his research. The causality of his discovery has been debated and researched ever since. Needless to say, avoiding sugar and its acidity is beneficial for our bodies. If cancer hates alkalinity, so much the better, if we decrease acidic sugar in our diet.

Feeding the cancer cell

As a positive person, you likely eat healthy, vegan, and nutritionally packed foot to provide you with the sustenance you need for your day. But what if your meal was accidentally feeding cancerous growth inside your body? Scientists explain that your balance of carbohydrates from sugars, both natural and added, might need to be changed.

Sugar, as a carbohydrate, breaks down for use as fuel for your cells. However, it also feeds cancer cells. Fat tissue stores excess sugar. Clearly, we don’t want to store food for cancer cells that want to grow and multiply. We want to eliminate any sugars stores in the fats of our bodies so that disease and cancer do not get an all-they-can-eat buffet.

Sugar stimulates cancer growth

Fermentation of sugar does not require oxygen, which is ideal for cancer cells. It helps them reproduce and multiply faster than if they did not have sugar. The scientists studying sugar’s effect on cancer growth found that sugar, just like in human children, causes hyperactivity for the reproduction and spreading of cancer cells. In other words, it dials it up to high speed and allows cancer to proliferate in the body.

Researchers found that the Warburg effect creates a vicious cycle through activation of early cellular growth pathways, like a roadmap for genes to build on. This later leads to enhanced fermentation of sugar and a lack of oxygen in the cellular environment, which stimulates increased growth of tumors.

Sugar changes cellular functions

Further research  examined sugar’s role in the growth of cervical cancers. Some of those who are affected by HPV (human papilloma virus) have an increased risk for developing cervical cancer. However, only some of the 200 genetic variants of HPV are associated with a higher risk of cancer growth.
These researchers found two different cellular mechanisms that are affected by sugar, which causes HPV to prompt cells to become cancerous and grow. The first connection between sugar and cancer growth is in the preference for the genetic expression of a process in the cell that enhances cellular growth under anaerobic or hypoxic conditions. The second connection between sugar and cancer is that it enhances the ability of cervical cancer cells to multiply due to changes in cell signaling.

Simply put, sugar makes it easier for a normal cell to mutate, which allows the mutated or cancerous cell to use sugar for energy even without oxygen. Sugar also affects the rate of cancer cell proliferation, causing it to multiply and spread faster.

Weighing the sweet against the risk

A diet rich is sugar can lead to other health problems, for example, obesity. The Body Mass Index (BMI) measures weight-to-body-height proportions for both men and women. A higher weight density or BMI associates with a higher sugar intake in general. Scientists  found that a higher BMI also associates, among other factors, with a higher risk for colon cancer.

As the body processes excess sugar in the bloodstream, a high glycemic index (GI) causes the pancreas to release insulin. Triglycerides are also formed to store excess sugars. High GI, insulin, and triglycerides are all associated with diets high in sugars and with a higher risk of colon cancer. Not surprisingly, exercise may help lower the risk of colon cancer. The researchers say that if the individual is sedentary, the risk for colon cancer is higher than if they are active.

One possible explanation for this is that exercise causes the body to burn some of the available sugar so that there is less of this acidic molecule in the bloodstream. Using up sugar during exercise also means that your body excretes less sugar as waste. As a result, less sugar reaches the colon.

What can we do to manage our sugar and cancer risk?

The Physicians give us some basic recommendations about sugar in the diet, especially when there is a low fiber content, also called a higher glycemic index. Whole, unprocessed, vegan foods are preferable for the diet of a positive thinking person. Whenever possible, choose these over cooked, processed, or packaged foods.

The less processed a food is, the less nutritional value it loses. Thus, the less environmental impact it has. It is also less likely that simple sugars have been added through processing. Sources of carbohydrates that are multi-grain are preferred over whole grain. Furthermore, whole grains trump bleached flour when it comes to protecting your health. Fiber helps offset some of the negative effects of sugar on cancer growth.


Think about changing some of your foods to reduce the total amount of simple carbohydrates. (These are often found in crackers, white bread, cookies, cereal, candy, and sweetened beverages).
