Chemists develop promising molecular tool to target cell immortality's underlying gears

One of the hallmarks of cancer is cell immortality. A Northwestern University organic chemist and his team now have developed a promising molecular tool that targets and inhibits one of cell immortality's underlying gears: the enzyme telomerase.

This enzyme is found overexpressed in approximately 90% of human cancer cells and has become an important subject of study for cancer researchers. Normal cells have the gene for telomerase, but it typically is not expressed.

"Telomerase is the primary enzyme that allows cancer cells to live forever," said Karl A. Scheidt, who led the research. "We want to short-circuit this immortality. Now we have designed a first-of-its-kind small molecule that irreversibly binds to telomerase, shutting down its activity. This mechanism offers a new pathway for treating cancer and understanding cellular aging."

Scheidt is a professor of chemistry in the Weinberg College of Arts and Sciences and a professor of pharmacology at Northwestern University Feinberg School of Medicine.

The big idea for the small molecule design came from nature. A decade ago, Scheidt was intrigued by the biological activity of chrolactomycin, which is produced by bacteria and has been shown to inhibit telomerase.

Scheidt and his team used chrolactomycin as a starting point in the design of their small molecules. They produced more than 200 compounds over the years, and the compound they call NU-1 was the most effective of those tested. Its synthesis is very efficient, taking fewer than five steps.

    NU-1 inhibits telomerase unlike anything that came before it. It does this by forming a covalent bond. Another advantage of NU-1 is that its molecular structure should enable scientists to add cargo, such as a therapeutic." Karl A. The study was published last week by the journal ACS Chemical Biology.

All human cells have telomeres, short DNA sequences that cap the ends of each strand of DNA. Their job is to protect our chromosomes and DNA. When a cell divides, the telomeres get shorter until they can no longer do their job. Natural cell death follows.

In contrast, cancer cells, with their heightened telomerase activity, become immortal by reversing the normal telomere shortening process. The enzyme telomerase copies telomeres over and over again, lengthening the telomeres. The result is unlimited cell division and immortality. The famous HeLa cells, isolated from the cervical cancer tissue of Henrietta Lacks in the 1950s, are still dividing.

Telomerase has been a target for cancer therapeutics research for decades. In 2009, three scientists received the Nobel Prize in Physiology or Medicine for their earlier research into telomeres and telomerase.




After developing their new compounds, Scheidt and his team initiated collaborations with Professor Stephen Kron at the University of Chicago and Scott Cohen at the Children's Medical Research Institute in Sydney to investigate the extra-telomeric role of telomerase inhibition.

The studies focused on how the new compounds interact with telomerase on a molecular level and how telomerase inhibition sensitizes cells to chemotherapies and irradiation. From this work, NU-1 rose to the top.

"By publishing this study, we are test driving this exquisite tool to see what it can do and to learn more about telomerase," Scheidt said. "We also are continuing to make it better."

The research was done in human cells. The next steps, Scheidt said, are to make more potent compounds and investigate them in animal models.

Foot and mouth disease virus to help treat pancreatic cancer

The dreaded pancreatic cancer has the lowest survival rates among all types of cancers. However, a group of scientists are using the foot and mouth disease virus to find a cure for this often lethal condition.

The research team from the Queen Mary University of London has identified a peptide, or protein fragment, taken from the foot-and-mouth-disease virus that targets another protein, called AvB6 (alpha-v-beta-6). This protein is found at high levels on the surface of the majority of pancreatic cancer cells.

The study was published in Theranostics.

Working jointly with Spirogen and ADC Therapeutics, the team have used the peptide to carry a highly potent drug, called tesirine, to the pancreatic cancer cells. When mice with pancreatic cancer tumours were treated with the drug and peptide combination, the tumours were completely killed.

"Foot-and-mouth-disease virus uses AvB6 as a route to infect cattle, as the virus binds to this protein on a cow's tongue. By testing pieces of the protein in the virus that attaches to AvB6, we've developed a route to deliver a drug specifically to pancreatic cancers," said John Marshall, lead researcher professor from the Cancer Research UK Barts Centre.

The team performed tests of the peptide/tesirine combination in both cells in the laboratory and in mice.

Mice that had AvB6-positive tumours were given a tiny dose of the peptide-drug combination three times a week, and this stopped the tumours growing completely. But when the dose was increased and given just twice a week, all tumours in mice that were AvB6 positive were completely killed.

From the experiment, scientists were excited to offer a completely new way of treating pancreatic cancer.

Prof Marshall explained "One advantage of targeting AvB6 is that it is very specific to cancer because most normal human tissues have little or none of this protein. So we're hopeful that, if we can develop this into an effective treatment for pancreatic cancer, it would have limited side effects."

The team now plan to further test the peptide and drug combination in more complex mice models, to determine if it can also impact on pancreatic cancer metastases, before moving to clinical trials.

Scientists compared brain scan results of people with a diagnosis of psychosis and people without the disorder as they were doing a memory exercise. In the people with psychosis, the researchers found clear differences in the activity patterns in their prefrontal, mediotemporal, and striatal areas of the brain — those that regulate decision making, learning and memory. But when these participants were given a single dose of CBD, their brain activity patterns were more similar to those of people without the disorder. 

The results provide insight into which part of the brain is affected by CBD and suggest CBD may curb symptoms of psychosis in the human brain.

The study was published Wednesday in Psychological Medicine.

Health Risks Associated With Daytime Naps

Who doesn’t love a good nap during the day? It feels like the perfect way to catch up on deficit sleep from the night, to recharge your batteries and refresh yourself. But while it may be tempting to take one for all the relaxation it offers, it could also indicate some major health disorder including Alzheimer's Disease, according to a latest research. The research found that Alzheimer’s first targets the brain cells that keep us awake during the day. This tends to make us sleepier during the day.  That means that if you are one of those who feel sleepy during the day you could be at risk of developing the neurological disease.

In September last year, another study,  said that those who feel sleepy during the day are nearly three times more likely to develop Alzheimer’s disease than those who have a good night’s sleep. Those who report being very sleepy during the day are more likely to have brain deposits of beta-amyloid — a protein that’s a hallmark for Alzheimer’s disease — years later. The finding, said that poor quality sleep could encourage this form of dementia to develop, suggesting that getting adequate night-time sleep could be a way to help prevent Alzheimer’s disease.

Heart disease: Another research associated daytime napping with the risk of developing cardiovascular disease (CVD) and death. The study found that people who slept for longer than the recommended duration of six to eight hours a day had an increased risk of dying or developing heart disease or stroke.

Insomnia: Daytime naps can disrupt your sleep at night causing sleeplessness. This is why those who experience sleep problems at night should make sure to get treatment for their problems instead of routinely trying to catch up on lost sleep during the day.

Novel drug may help tackle treatment-resistant cancers

A new class of drug that has the potential to help cancer patients, especially those suffering from breast cancer and who no longer respond to existing therapies has been discovered.

Breast cancer patients frequently become resistant to existing treatments, leading to the disease becoming fatal.

Early lab-based tests of a drug named ICEC0942 were successful in targeting resistant breast cancers and indicated minimal side effects.

“Treatment-resistant tumours represent a significant threat for patients, as once a cancer stops responding to treatments there is increasingly little clinicians can do,” said a Professor.

“Drugs such as these could help to shift the balance back in favour of the patients, potentially providing a new option to patients for who existing treatments no longer work,” the Prof. added.

The findings, published in the journal, noted that the drug targets an enzyme called CDK7 — involved in directing cells through their life-cycle, which consists of growth, DNA replication and cell division.

CDK7 is also involved in the process of transcription — a vital step in gene expression — the creation of proteins to carry out cell functions.

Particular cancers, such as treatment-resistant breast cancers, have a unique dependence on transcription, meaning targeting CDK7 may be particularly effective.

By inhibiting transcription, ICEC0942 shuts down the ability of the cancer to spread.

In addition, cancers such as acute myeloid leukaemia and small-cell lung cancer are particularly transcription-dependent.

Therefore, ICEC0942 drug may also work well for these too, especially where they have become resistant to other treatments, the researchers suggested.

Skipping Rope Is A Better Workout Than Jogging

Good news for all those who have been having a tough time to make room for high intensity work out. A skipping rope is all you need to make up all the cardio sessions you have been missing out on. According to a latest study, skipping could prove to be the best full body workout. The uncomplicated workout takes minutes and targets all your muscles, at the same time it is not too tiring or harsh.

The study, published in a Journal says, skipping is one of the most effective cardio exercises. The exercise also resulted in better cardiovascular health.

For the study, the scientists compared jogging and skipping and found the latter to be more effective. The study that went on for six weeks, monitored the two physical activity. The findings revealed that 10-minute jump-rope sessions a day were more efficient than 30-minute jogging sessions. Participants demonstrated improvement in their cardio-vascular health.

Jumping up and down, proved to be more efficient for the target muscles than hitting the pavement. The research also reveals that skipping burns more calories per minute and engages more muscles than rowing or swimming.
