Illama-derived antibodies may block coronavirus entry into host cells

Molecules derived from the immune system of the South American mammals illamas may block the entry of the novel coronavirus into host cells, according to a study which could lead to a potential avenue for COVID-19 treatment. Researchers, including those from The University of Texas at Austin in the US, have linked two copies of a special kind of antibody molecules produced by the llama immune system to create a new type that binds tightly to a key protein on the novel coronavirus, SARS-CoV-2.

According to the study, published in the journal Cell, this protein, called the spike protein, allows the coronavirus to break into host cells. The findings, according to the researchers, indicate that the Illama-derived antibody may block viruses with this spike protein from infecting cells cultured in the lab. "This is one of the first antibodies known to neutralise SARS-CoV-2," said study co-author Jason McLellan from UT Austin.

The scientists said they are preparing to conduct preclinical studies in animals such as hamsters or nonhuman primates, with the goal of developing a treatment that would help people soon after infection with the virus. While vaccines have to be given a month or two before infection to provide protection, the researchers said antibody therapies can achieve this immediately.

"With antibody therapies, you're directly giving somebody the protective antibodies and so, immediately after treatment, they should be protected," McLellan said. "The antibodies could also be used to treat somebody who is already sick to lessen the severity of the disease," he added. According to the researchers, this would be especially helpful for vulnerable groups such as elderly people, who mount a modest response to vaccines, meaning that their protection may be incomplete.

The scientists explained that the strategy can be made to work since the immune system of illamas produce antibodies similar to that of humans. When llamas' immune systems detect foreign invaders such as bacteria and viruses, these animals produce two types of antibodies. One of these is similar to human antibodies, and another that's only about a quarter of the size, the researchers said.

They added that the smaller ones, called single-domain antibodies or nanobodies, can be nebulised and used in an inhaler. "That makes them potentially really interesting as a drug for a respiratory pathogen because you're delivering it right to the site of infection," said Daniel Wrapp, study co-author from UT Austin.

The scientists extracted the antibodies for the study from a 4-year-old illama, Winter, living on a farm in the Belgian countryside along with approximately 130 other llamas and alpacas. In 2016, when Winter was about 9 months old, the researchers were studying two earlier coronaviruses -- SARS-CoV-1 and MERS-CoV. In a process similar to humans getting shots to immunise them against a virus, the researchers injected the illama with stabilised spike proteins from the coronaviruses over the course of about six weeks.

They then collected a blood sample and isolated the antibodies bound to each version of the spike protein. According to the researchers, one of these antibodies, VHH-72, showed promise in stopping a virus with spike proteins from SARS-CoV-1 from infecting cells cultured in the lab. "There wasn't a big need for a coronavirus treatment then. This was just basic research. Now, this can potentially have some translational implications, too," Wrapp said.

The scientists then engineered a new antibody that showed promise for treating the current SARS-CoV-2. According to the researchers, the engineered antibody involved linking two copies of VHH-72 that bound more effectively to the novel coronavirus spike protein. They said the engineered protein can bind strongly to viruses displaying spike proteins from SARS-CoV-2 in cell cultures, adding that this is the first known antibody that neutralise both SARS-CoV-1 and SARS-CoV-2.

Oranges- cold, cholesterol !

Oranges have been traditionally used to ward off colds, but are also great to lower cholesterol. The vitamin C in oranges has been hyped as one of the best home remedies. But in recent years, researchers have found oranges are beneficial for reducing cholesterol. A dietitian says “Oranges are rich in pectin, a soluble fiber that creates a gel-like substance inside your body that binds to bad cholesterol and carries it out. Eating a diet rich in soluble fiber can help to reduce unhealthy LDL cholesterol levels as well as total cholesterol levels.”

Implantable device using light to treat bladder problems developed

A team of neuroscientists and engineers has developed a soft, implantable device that can detect over-activity in the bladder and then use light from tiny, bio-integrated LEDs to tamp down the urge to urinate.

In a minor surgical procedure, the researchers implant a soft, stretchy belt-like device around the bladder. The researchers also inject proteins called opsins into the animals' bladders. The opsins are carried by a virus that binds to nerve cells in the bladder, making those cells sensitive to light signals. This allows the researchers to use optogenetics, the use of light to control cell behavior in living tissue, to activate those cells.

Using blue-tooth communication to signal an external hand-held device, the scientists can read information in real time; and using a simple algorithm, they detect when the bladder is full, when the animal has emptied its bladder, and when bladder emptying is occurring too frequently.

"When the bladder is emptying too often, the external device sends a signal that activates micro-LEDs on the bladder band device, and the lights then shine on sensory neurons in the bladder. This reduces the activity of the sensory neurons and restores normal bladder function," said  a professor of anesthesiology  and one of the study's senior investigators.

The researchers believe a similar strategy could work in people. They expect to test similar devices in larger animals. They also believe the strategy could be used in other parts of the body, say treating chronic pain, or using light to stimulate cells in the pancreas to secrete insulin.

But, one hurdle involves the viruses used to get light-sensitive proteins to bind to cells in organs.

"We don't yet know whether we can achieve stable expression of the opsins using the viral approach and, more importantly, whether this will be safe over the long term," the researcher said.

Reasons Why Broccoli Is Called A Superfood !

Everyone knows that eating veggies is incredibly good for your health, but if we're really honest with ourselves, a plate of broccoli has nothing on a dish full of curly fries! However, a new study  has just discovered yet another reason why we should eat our veggies, which is particularly important for those of us who suffer from digestive issues.

When working with lab mice, the team discovered that their mice could better tolerate digestive issues after eating broccoli than mice who weren't fed any veg. According to a professor of agricultural sciences, "there are a lot of reasons we want to explore helping with gastrointestinal health and one reason is if you have problems, like a leaky gut, and start to suffer inflammation, that may then lead to other conditions, like arthritis and heart disease."
The researchers believe that broccoli was effective thanks to the way certain compounds it contains bind to stomach receptors called Aryl hydrocarbon receptors, or AHRs. When cruciferous vegetables such as broccoli are broken down in the stomach, one of the resulting compounds is called indolocarbazole, or ICZ, which in turn has been found to bind to AHR.
"When ICZ binds to and activates the Aryl hydrocarbon receptor (AHR) in the intestinal lining, it aids in maintaining a healthy balance in the gut flora and immune surveillance, and enhances host barrier function," according to the research team. "This may help prevent diseases, such as various cancers and Crohn's Disease, caused by inflammation in the lining of the gut."
In their study, the researchers worked with two types of mice which were genetically different. One type had an enhanced ability to bind ICZ to AHR, while the other type had a reduced ability to do so. All mice were placed on a diet which contained either 15% broccoli or which contained no broccoli at all. The results showed that after being given substances to cause digestive issues, only those mice that had consumed broccoli managed to escape the uncomfortable symptoms.
To consume an equivalent amount of broccoli, humans would need to eat 3 and a half cups each day, which may definitely be quite a lot for some people. However, the Prof. claims that there may be alternative ways of getting the ICZ-to-AHR binding benefits.
"Now, three and a half cups is a lot, but it's not a huge amount, really," he claims. "We used a cultivar — or variety — with about half the amount of this chemical in it, and there are cultivars with twice as much. Also, Brussels sprouts have three times as much, which would mean a cup of Brussels sprouts could get us to the same level."
The researchers will also investigate ways to deliver the health benefits of broccoli to people who need to avoid high fiber levels in their diets because of digestive issues.

Scientists develop affordable tool to detect bacteria in food, water

A team of scientists has reportedly developed a new low-cost method for detecting bacteria in food or water samples.

A team  along with a food scientist, developed the tool which could be of use to cooks for fresh fruits and vegetables, once available commercially.

An analytical chemist and expert in detection methods for food contamination, said, "People around the world cook their vegetables before eating, but here in the U.S. more and more people like to eat these foods raw. This gave us the idea that a quick test that can be done at home would be a good idea."

Insisting on the importance of microbial contamination research, she added, "It has been a problem for a long time, but it is now the number one concern for food safety in the US".

The food researchers reported on their two-step method - one chemical, one optical - in two papers this year. They designed a reliable and sensitive bacteria-detecting chip that can test whether food or liquid samples carry a bacterial load. The chip, used with a light microscope for optical detection, relied on what the researcher called a "capture molecule," 3-mercaptophenylboronic acid (3-MBPA) that attracts and binds to any bacteria.

According to the accompanying report, the chemical detection method, "surface-enhanced Raman spectroscopy" or SERS, relied on silver nano-particles. The techniques are now in the patenting process.

Over the past few months, the optical detection method was adapted for possible home use. With the smartphone microscope adapter that is widely available online, it could be used to detect contamination in food items.

It would require samples of water, juice or mashed vegetable leaf to place the chemical-based chip in for detection. the researcher reported that a summer high school student had developed a simple app for smart phones that can visually detect bacteria in samples containing the chip.

The researcher called it just a "beginning of the work" and is hoping to get appropriate funding to continue this practical application.

New anti-ageing protein may combat obesity, cancers

Scientists have revealed the three-dimensional (3D) structure of an anti-ageing protein, that may not only increase human lifespan but also help develop therapies for age-related diseases such as diabetes, obesity and cancers.

The Klotho proteins -- alpha and beta -- named after the Greek goddess Clotho, who spun the thread of life, play an important role in the regulation of longevity and metabolism.

Both work in tandem with molecules called endocrine FGFs, which regulate metabolic processes in tissues and organs including the brain, liver and kidneys, among other organs.

In the study,  the team found that beta-Klotho is the primary receptor that binds to FGF21 -- a key hormone produced upon starvation.

FGF21 has the potential for boosting insulin sensitivity and glucose metabolism, causing weight loss.
This new understanding of beta-Klotho and FGF21 can guide the development of therapies for conditions such as Type 2 diabetes in obese patients, the researchers said, in a paper published .

"Like insulin, FGF21 stimulates metabolism including glucose uptake. In animals and in some clinical trials of FGF21, it shows that you can increase burning of calories without changing food intake, and we now understand how to improve the biological activity of FGF21," said a researcher.
A new variant of FGF21 with 10 times higher potency and cellular activity was also unveiled.

By developing drugs that enhance the pathway, the researcher said, researchers can target diabetes and obesity.

Conversely, using agents that block the pathway, they also hope to explore therapies for conditions such as liver cancer and bone diseases, among others.

"The next step will be to make better hormones, make new potent blockers, do animal studies, and move forward," the researcher said.
