Study shows how extracellular vesicles help melanoma cells to spread

A new study from Finland sheds fresh light on how melanoma cells interact with other cells via extracellular vesicles they secrete. The researchers found that extracellular vesicles secreted by melanoma cells use the so-called hedgehog signaling pathway to intensify the malignant properties of the cells they are targeting. The discovery can help in the development of better treatment and diagnostics for melanoma. Published in Cellular and Molecular Life Sciences, the study was carried out in collaboration between researchers from the University of Eastern Finland and the University of Helsinki.

Many of the mechanisms regulating the function of our body have remained unchanged throughout evolution. This means that the same genes regulate the development and function of all multicellular animals. One example of such gene survival is the so-called hedgehog signaling pathway. The gene family associated with this signaling pathway gets its unique name from a mutation it caused in fruit fly larvae, making them look spiky, just like a hedgehog. The most common of the three hedgehog genes found in mammals, the Sonic hedgehog gene, was aptly named after the famous video game character. In mammals, members of the hedgehog gene family are essential regulators of fetal development, but they are also associated with stem cell division in later stages of development and even in adults. Recent studies suggest that their expression is also associated with many different types of cancer, including skin cancers. Melanoma is the most severe form of skin cancer, and it is becoming more and more common.


Extracellular vesicles carry signals between cells - and they can also carry packaged drugs

Traditionally, signaling in the body is thought to take place via freely circulating signals, such growth factors and hormones. According to a more recent view, however, it is believed that some signals are packaged for transportation to protect them against breakage and to ensure their delivery to the correct address. Extracellular vesicles are small, bubble-like packages made of cell membrane, serving as natural carriers of signals in the body. They regulate the function of the body already during the fetal stage, via breast milk and also later in life as our tissue regenerates. Cancer cells, too, use extracellular vesicles to deliver signals and to modify their environment, making it favorable to growth. Extracellular vesicles can also be used as diverse carriers of drugs to combat diseases and to repair tissue damage - and this is why they are being studied so actively at the moment.


In the newly published study, the researchers discovered a new link between extracellular vesicles and hedgehog molecules. They found that vesicles secreted by melanoma cells intensify the malignant properties of the cells they are targeting, such as division and spreading, via the hedgehog signaling pathway. The researchers used cultured human melanoma cells and normal skin cells, confirming their findings from the cell culture by analyzing tissue samples from patients with melanoma.

It is quite a coincidence that these signal-carrying vesicles originate from cells that are also known as hedgehog cells due to their microscopically small, spike-like protrusions. These protrusions, however, don't have anything to do with fruit fly larvae; instead, they are typically found in cells that are active in producing hyaluronan, the most common sugar molecule in the extracellular matrix. Hyaluronan also plays a key role in vesicle mediated signaling, as hyaluronan found on the surface of vesicles protruding from the cell surface facilitates their binding to the target cell."  Docent Kirsi Rilla, University of Eastern Finland

The hedgehog signaling pathway holds great promise as a target for drug therapy in melanoma and other cancers. The regulation mechanism now discovered by the researchers can be made use of in the development of better diagnostics and drug therapy for patients with melanoma.

Docent Rilla's research group has been studying the biology of extracellular vesicles since 2013.

Placenta health depends on maternal exercise

Do you know that exercising during pregnancy can keep your placenta healthy? A recent study shows that maternal exercise is important for fetal development in obese mothers.

Over 1.9 billion adults worldwide  are overweight or obese, and by 2-25 this is projected to increase to 2.7 billion. Obesity significantly raises the risk of developing 11 different types of cancer, stroke, type 2 diabetes, heart disease, and non-alcoholic fatty liver disease.


The placenta performs nutrient and oxygen exchanges between the mother and her foetuses, and placental function is impaired due to maternal obesity, which is likely to later nutrient and oxygen delivery to the foetus.


Exercise during pregnancy is known to be beneficial for both maternal health and fetal development. This study found that exercise improved the function of the placenta, and also the metabolism of the mother. Importantly, they found that the babies were both born larger ( which is frequently associated with obesity and metabolic diseases in later life, when the obese mothers exercised.


This study was conducted on animal models. In this study, female mice were fed a healthy diet ( 10% energy from fat) or a high-fat diet to become obese and them mated. Each of the maternal groups was further divided into 2 sub-groups- those that did and did not perform exercise during gestation.
Mice were exercised from 0-18.5 days of pregnancy ( the term is 20.5 days) and placenta function and maternal and fetal changes were analysed.


Further studies will focus on identifying mechanisms explaining the beneficial effects of exercise on placental development of obese mothers. Researchers will define the possible role of chemicals secreted during exercise, on blood vessel development in the placenta, which is critical for the delivery of nutrients and oxygen to fetuses, as well as their long-term impacts on the health of next generation.


The senior author of the research said, understanding how maternal exercises might help prevent offspring from becoming obese or developing metabolic diseases will help us to best guide mothers so they can ensure their babies are as healthy as possible.

Australian researchers discover how a baby's sex is determined

While it is known that the sex of a baby is determined by its chromosome make-up at conception, new research has identified a "regulator" that increases or decreases the activity of genes which decide if we become male or female.

"These regulatory segments of DNA are called enhancers," said lead author of the study.

"If these enhancers that control testis genes are disrupted it may lead to a baby being born with a disorder of sex development," Sinclair said. 

While an embryo with two X chromosomes will become a girl, an embryo with an X-Y combination results in a boy. But the Y chromosome carries a critical gene, called SRY, which acts on another gene called SOX9 to start the development of testes in the embryo, explained a researcher.

"High levels of the SOX9 gene are needed for normal testis development," he added. 

This study,  sought to understand how the SOX9 gene was regulated by enhancers and whether disruption of the enhancers would result in disorders of sex development.

"We discovered three enhancers that, together ensure the SOX9 gene is turned on to a high level in an XY embryo, leading to normal testis and male development," he said.

"Importantly, we identified XX patients who would normally have ovaries and be female but carried extra copies of these enhancers, (high levels of SOX9) and instead developed testes," Sinclair said.
"In addition, we found XY patients who had lost these SOX9 enhancers, (low levels of SOX9) and developed ovaries instead of testes," Sinclair added.

Croft said human sex reversal such as seen in these cases is caused by gain or loss of these vital enhancers that regulate the SOX9 gene.

Poor nutrition during pregnancy may advance menopause

While it is known that inadequate nutrition during fetal development can cause diabetes and heart disease, a new study warns that it could also lead to early menopause at age younger than 45. Prenatal malnutrition was also associated with a higher risk of premature ovarian failure. Natural menopause is a milestone of ovarian aging that results in the end of a woman's reproductive years.

Conversely, an early menopause is associated with increased risk of heart disease, osteoporosis, depression, and memory changes and changes in vaginal and sexual health, according to researchers. "The findings showed that food deprivation during early fetal life affects how long the future ovaries function," said a researcher.. 

Although several studies have investigated the association between malnutrition exposure in early life and risk of various metabolic diseases in adulthood, the association with reproductive ageing was not evaluated.

This new study, involved nearly 2,900 Chinese women and specifically sought to address the effect of early life exposure to malnutrition on age at menopause.

It has already been documented that the development of the hypothalamic-pituitary-gonadal axis during the fetal stage plays a critical role in adulthood reproductive health, the researchers said.

Nutritional benefits of Capsicum / Bell peppers

Capsicum is known by a variety of names such as Bell Peppers, Paprika, Cayenne, and Chilies. They come in various colours like green, red and yellow. It is an excellent source of Vitamin A, Vitamin C, beta-carotene and other carotenoids, such as lutein, beta-cryptoxanthin, and zeaxanthin.

1.Burns more Calories:
Red bell peppers help in activating thermogenesis and increase the metabolic rate. Capsaicin which is responsible for increasing the hotness of other peppers is found in bell peppers in very small amounts. Thus, they have a mild thermogenic action that increases the metabolism without increasing the heart rate and blood pressure unlike hot peppers. Hence, they can support weight loss.

2. Good For Eye-sight-
 Vitamin A which is necessary for healthy vision is present in abundance in capsicum. The role of vitamin A in strengthening the immune system along with its antioxidant properties makes capsicum a must included food in your diet.

Red bell peppers are rich in lycopene, thus making them excellent for a healthy heart whereas green bell peppers are a good source of cholesterol lowering fiber.
