The Ins and Outs of Collagen

Collagen is a hard, insoluble, and fibrous protein that makes up one-third of the protein found in the human body. It acts as a supporting structure and anchors cells to each other to provide strength and elasticity. There are at least 16 different types of collagen, but 80-90% of them belong to types 1, 2, and 3. Each type has a different structure and functions.

What Does Collagen Do?  
Collagen is secreted by various cells, but mostly by connective tissue cells. It’s found in the extracellular matrix, which is an intricate network of macromolecules that determines the physical properties of body tissues. A macromolecule is a molecule that contains a large number of atoms. 
 
In the dermis, or the middle layer of the skin, collagen helps to form a fibrous network of cells known as fibroblasts, upon which new cells grow. It also plays a role in replacing and restoring dead skin cells. Some collagens act as protective layers for delicate organs such as the kidney. 
 
With age, the body produces less collagen, resulting in the formation of wrinkles, and weaknesses in joint cartilage. Women experience a dramatic reduction in collagen synthesis straight after menopause.  
 
Uses: Medical and Cosmetic
Collagen is resorbable which means it can be broken down, converted, and absorbed back into the body. Furthermore, it can also be formed into compacted solids or lattice-like gels. Its diverse range of functions, and the fact that it occurs naturally, makes it clinically versatile and suitable for a range of medical procedures.

Skin Fillers
Collagen injections can improve the contours of the skin and help fill out depressions. Fillers that contain collagen can be used to remover wrinkles and lines from the face. They can also help improve scars, as long as they do not have a sharp edge.

Wound Dressing 
Collagen can help to heal wounds by attracting new skins cells to the wound site. It promotes healing and provides a new platform for tissue growth. 
 
Collagen dressings can help heal: 
• Chronic wounds that do not respond to other treatments 
• Necrotic or rotting wounds 
• Wounds that expel bodily fluids such as urine or sweat 
• Granulating wounds, on which different tissues grow 
• Partial and full-thickness wounds 
• Second-degree burns 
• Sites of skin donation and skin grafts 
 
Guided Tissue Regeneration 
Collagen-based membranes have been used in periodontal and implant therapy to promote the growth of specific types of cells. In oral surgery, collagen barriers can prevent fast-growing cells around the gum from migrating to a wound in a tooth. This preserves a space, allowing tooth cells the chance to regenerate. 
 
Vascular Prosthetics  
Collagen tissue grafts from donors have been used in peripheral nerve regeneration, in arterial reconstruction, and in vascular prostheses. While collagen prostheses are compatible with the human body, some have been found to be thrombogenic, or likely to cause coagulation of the blood.  
 
Treatment of Osteoarthritis

Collagen supplements may help treat osteoarthritis. A study carried out back in 2006 found that supplements containing collagen helped decrease painful symptoms and improve joint function in people with osteoarthritis. As the supplement is absorbed, collagen accumulates in the cartilage, helping to rebuild the extracellular matrix. 
 
Nutrients That promote Collagen Formation 
 
• Proline: In egg whites, cheese, soy. Cabbage, and meat. 
 
• Anthocyanidins: In blueberries, cherries, blackberries, and raspberries. 
 
• Vitamin C: In strawberries, oranges, broccoli, and peppers. 
 
• Copper: In shellfish, red meat, nuts, and some drinking water.

What Damages Collagen? 
Some factors can deplete the levels of collagen in the body. Avoiding them could keep the skin healthier for longer. 
 
High Sugar Consumption 
A diet high in sugar increases the rate of glycation, a process where blood sugars attach to proteins to form new molecules called advanced glycation end products (AGEs). These damage nearby proteins and can make collagen brittle, dry, and weak. 
 
Smoking
Many of the chemicals found in tobacco smoke damage both collagen and elastin in the skin. Nicotine also narrows the blood vessels in the outer layers of the skin, which reduces the amount of nutrients and oxygen that can be delivered to the skin. 
 
Sunlight 
UV rays cause collagen to break down more rapidly, damaging collagen fibers and causing an abnormal buildup of elastin. When the collagen in the dermis is damaged by UV rays, the skin rebuilds incorrectly, forming wrinkles. 
 
The Aging Process  
Collagen levels decrease over time and there is nothing that can be done to prevent this. Avoiding tobacco, excess sun exposure, and following a healthy diet and exercise regime can help reduce visible aging and protect collagen, keeping the skin, muscles, bones, and joints healthy for longer.

Scientists revive brain funciton in dead pigs

Scientists have managed to restore cell function in the brains of pigs hours after they died, in a breakthrough that experts said threw into question the very notion of what makes animals of even humans--alive.

Authors said their research could one day be used to help treat victims of heart attacks and strokes and unravel the mysteries of brain trauma.


In human and large mammal brains, cells crucial for neural function begin to degrade as soon as blood supply is cut- a process long considered to be irreversible.


But the findings of the trial, show that the brains of pigs can have their blood flow and cell function restored even hours after death.


The team used the brains of 32 pigs that had been slaughtered for food and discarded, without blood or glucose flow for 4 hours each.


Then, using a tissue support system that pumps a liquid designed to mimic blood through organs, they rehydrated the brains for a further 6 hours.


Some brains received the patented life-support solution and others were given a control substance.
The results were astonishing: the brains that recived the artificial blood had basic cell function restored. Their blood vessel structure was revived, and the team even observed some localised processes- including in synapses and immune responses- flickering back to life.


Those that received the control solution turned within hours to what researchers described as something a bit like yoghurt.


I was tremendously surprised, a sresearcher and lead study author said.


We were surprised how well the structure was preserved. We found that cell death is reduced, which is very exciting and promising.


The main conceptual finding is this: cell death in the brain occurs across a longer time period than we previously thought , added the author.


The team stressed that they saw no higher level functional activity, such as electric signalling associated with consciousness in the revived brains.


That is a clinical sign that the brain is alive, and we never found it, he said. This is not a living brain, it is a cellularly active brain.


Had any such activity been detected, the team said they would have had to deliver anaesthetic to the brains to null any pain- despite then having been dead for hours.


If some activity shows up that indicated consciousness, we would have to stop the experiment, said a researcher.


This would be unique: inducing consciousness in an organ that's not connected to any living being.
The findings suggest that scientists may have under appreciated the brain's capacity for self-restoration after a patient is declared brain dead.


But for experts invited to comment on the study, it raised deeper philosophical and ethical questions.
Writing in a linked editorial, a Prof. of law and philosophy said that the study throws into question long-standing assumptions over what makes an animal or a human alive.


She said researchers had inadvertently created an ethical grey area where the pigs used were not alive, but not completely dead.


A Prof. of medical ethics said that the study could have vital implications for future brain research.
This research reminds us that death is less an event, and more of a process that occurs over time, he said.


Cells within the human organism may be alive for some period of time after the person has died.


A researcher said the team planned to use their support system on dead brains for longer periods of time in order to see how the cells react.


We could potentially get some insight into how these brains could be saved, he said.

New Blood Test Can Detect Cancer in the Body in Just 10 Minutes

Wonderful health news is on the horizon in the world of cancer diagnosis. Researchers have developed a test that can detect the presence of cancer in the DNA within 10 minutes.

Scientists have homed in one what is being called a “universal cancer biomarker” that is easily identifiable with an inexpensive, portable test that could someday be accessed with a mobile phone.

The simple blood test focuses on finding cancerous cells in the DNA after they have died off and begun circulating in the bloodstream. It’s been discovered that these particular cells have a unique structure that differs from normal, healthy cells, which is marked by clusters of methyl groups.

What researchers have found is that those clusters – exclusive to cancer cells – cling easily to gold. Therefore, the test utilizes nanoparticles of gold for detection. One drop is all it takes and if cancer is present, the nanoparticles change color.

No microscope is needed. So far, 200 samples have been tested for human cancer types and healthy cells. The test has had a 90% accuracy rate, giving researchers hope that this will lead to early detection and as a result, earlier treatment.

The majority of cancer deaths are due to late stage detection. With this new technology, who knows how many lives can be saved.


This test costs less than typical biopsies used to diagnose cancer and although it’s not yet available to the medical community, it can be a real game-changer.

Altered DNA linked with psychiatric disorders identified

Brain scans have revealed a genetic mutation that affects the structure, function and chemistry of the brain, and thus increases the risk of major psychiatric disorders like schizophrenia, bipolar disorder and depression, says a research.
The findings showed that the people with the DISCI mutation caused a part of one chromosome to swap places with another as well as lead to changes in the structure of their brain. 


 These changes were associated with a significant increase in the risk of psychiatric illness, the researchers said.

"The study confirms and extends the genetics of DISC1, and shows how similar genetic effects can increase the risk of major mental illnesses," said Stephen Lawrie, Professor at the University of Edinburgh in Britain.

Further, the carriers of these mutations also had lower levels of glutamate -- a chemical in certain areas of their brain which can lead to schizophrenia, according to previous studies.

The DISC1 mutation was first identified in a Scottish family that showed unusually high rates of major psychiatric disorders. 


Scientists have been studying generations of the family for 40 years but this is the first time they have scanned their brains, said the paper appearing in the journal Schizophrenia.

Fat mass in cells expands with disuse

Researchers have claimed that nutrition is not the only factor driving obesity, asserting that the mechanics of “cellular expansion” plays a primary role in fat production, reports ANI.
To understand how obesity develops, Prof. Amit Gefen, Dr. Natan Shaked and Ms. Naama Shoham of Tel Aviv University’s Department of Biomedical Engineering, together with Prof. Dafna Benayahu of TAU’s Department of Cell and Developmental Biology, used state-of-the-art technology to analyze the accumulation of fat in the body at the cellular level.
Prog Gefan said that they wanted to find out why a sedentary lifestyle results in obesity, other than making time to eat more hamburgers, asserting that they found that fat cells exposed to sustained, chronic pressure — such as what happens to the buttocks when you’re sitting down — experienced accelerated growth of lipid droplets, which are molecules that carry fats.
He said that contrary to muscle and bone tissue, which get mechanically weaker with disuse, fat depots in fat cells expanded when they experienced sustained loading by as much as 50 per cent.
The researchers discovered that, once it accumulated lipid droplets, the structure of a cell and its mechanics changed dramatically.
Using a cutting-edge atomic force microscope and other microscopy technologies, they were able to observe the material composition of the transforming fat cell, which became stiffer as it expanded.
