FDA Approves First Therapy for Tumor-Induced Osteomalacia


The U.S. Food and Drug Administration approved Crysvita (burosumab-twza) injection to treat patients age two and older with tumor-induced osteomalacia (TIO).

 The Food and Drug Administration granted approval of Crysvita to Ultragenyx Pharmaceutical Inc.
 It is a rare disease that is characterized by the development of tumors that cause weakened and softened bones. The tumors associated with TIO release a peptide hormone-like substance known as fibroblast growth factor 23 (FGF23) that lowers phosphate levels.

Treatment for TIO focuses on identifying and removing the tumor that causes the disease. However, when that is not possible, Crysvita can help increase the levels of phosphate in the blood," said Theresa E. Kehoe, M.D., acting director of the Division of General Endocrinology in the FDA's Center for Drug Evaluation and Research. "As the first Food and Drug Administration-approved therapy to treat this debilitating disease, today's action is an important step in finding treatment options for patients living with TIO whose tumor cannot be found or removed."

FGF23 regulates levels of phosphate, an electrolyte that plays important roles in bone maintenance, energy production by cells and nerve function. When there is not enough phosphate in the body, bones begin to soften and weaken, causing osteomalacia (marked softening of bones).

The safety and efficacy of Crysvita were evaluated in two studies that together enrolled 27 adults with TIO. In both studies, patients received Crysvita every four weeks. For the first study, half of patients achieved normal phosphate levels through week 24 and maintained normal or near normal phosphate levels through week 144. In the second study, 69% of participants achieved normal phosphate levels through week 24 and maintained normal or near normal phosphate levels through week 88. The results of bone scans for patients in the first study also suggested healing of the bone lesions related to osteomalacia (softening of the bones).

Hypersensitivity reactions such as rash and hives have been reported in patients who took Crysvita. If serious hypersensitivity reactions occur, patients should stop taking Crysvita and talk with their health care provider about further medical treatment. Additionally, higher than normal levels of phosphorus may be associated with an increased risk of nephrocalcinosis (a disorder that occurs when too much calcium is deposited in the kidneys). 

The most common side effects reported in adults with TIO taking Crysvita were tooth abscess (nfection), muscle spasms, dizziness, constipation, injection site reaction, rash and headaches.
Patients taking oral phosphate or active vitamin D, those who have serum phosphate levels within or above the normal range for their age, and patients with severe kidney impairment or end stage renal disease should not take Crysvita. 

Crysvita is also Food and Drug Administration -approved to treat 

adults and children six months and older with X-linked 

hypophosphatemia, which causes low levels of phosphate in the 

blood and leads to impaired bone growth and development in 

children and teenagers.

Try to indulge in some form of physical activity for minimum 30 minutes on a daily basis. The best effective and simplest to do exercise is brisk walking," she says.

To ensure fitness on-the-go, you can take some simple measures like taking stairs instead of escalators, park your vehicle a little far from where you usually park and then walk to cover the distance, and take a couple of minute walking break from your monotonous office sitting hours.


Cholesterol: Other foods that can help in improving your numbers


1. Omega-3 fatty acids

Fatty fish or foods rich in omega-3 fatty acids should be included in cholesterol diet. Omega-3 fatty acids can reduce triglycerides, which is a type of fat in blood. Omega-3 fatty acids can also help in normalising blood pressure and reduce risk of blood clots. Tuna, mackerel, herring, salmon, trout, walnuts and chia seeds are examples of foods rich in omega-3 fatty acids.


2. Olive oil

Researchers discover new therapy target for drug-induced liver failure

A new research from a mouse study has suggested that treatments that increase levels of the protein thrombospondin-1 could help the liver recover from an overdose of acetaminophen.

According to the research presented at a meeting, Acetaminophen, a commonly used pain reliever, and fever reducer is the leading cause of quickly developing, or acute, liver failure in the United States.

“The only treatment available now for acetaminophen overdose must be administered early before significant liver injury occurs,” said the researcher.

“We are working to identify new drug targets to improve treatment options and reduce the need for liver transplantation, which is often the only option for patients with acute liver failure due to drug toxicity,” he added.

Researchers found that mice with acute liver failure from acetaminophen toxicity had higher levels of thrombospondin-1 than mice with normal liver function.

When they administered acetaminophen to genetically modified mice that lack thrombospondin-1, the liver injury and loss of liver function was more severe compared to unmodified mice.

The researchers also observed impaired liver regeneration and increased cell death in the mice without thrombospondin-1.

“Our study is the first to investigate thrombospondin-1 during acetaminophen toxicity. We were able to identify cell communication pathways that do not work properly during acetaminophen-induced liver injury and thus lead to worse outcomes,” said the researcher.

Next, the researchers plan to give mice different drug treatments that manipulate thrombospondin-1 to find out which treatment might be suited for potential clinical studies.They also plan to examine patients with acetaminophen-induced acute liver failure to determine if their thrombospondin-1 findings from mice hold true in people.

What women eat during pregnancy can modulate risk of ADHD in children

According to a new study, the risk of children  developing attention deficit hyperactivity disorder (ADHD) symptoms can be modulated by  mother’s diet during  while they are pregnant. The study,  had examined samples of umbilical cord plasma to calculate the levels of omega-6 and omega-3 which tend to reach the foetus. According to statistics, a higher omega-6: omega-3 ratio can be linked  to a higher risk of the ADHD symptoms at the age of 7.

Omega-6 and omega-3 can be termed as the  long-chain polyunsaturated fatty acids which have  a crucial role in the function and architecture of the central nervous system, mainly in the later stages of gestation.

These two fatty acids compete for incorporation into cell membranes and are primarily obtained through diet. Since omega-6 and omega-3 have opposing physiological functions, the former promotes systemic pro-inflammatory states, while the latter promotes anti-inflammatory states, a balanced intake of these two fatty acids is important. Previous research had shown that children with ADHD symptoms have a higher omega-6: omega-3 ratio.

The authors studied data from 600 children living in four Spanish regions (Asturias, Basque Country, Catalonia, and Valencia) who are participating in the INMA Project. They analysed umbilical cord plasma samples and data from questionnaires completed by the children`s mothers. The ADHD symptoms were assessed using two standard questionnaires: the first completed by the children`s teachers at the age of four years, and the second by their parents at the age of seven years.

The results showed that, at the age of seven years, the number of ADHD symptoms increased by 13 per cent per each unit increase in the omega-6: omega-3 ratio in umbilical cord plasma. The study analysed the number of symptoms in the children who met the diagnostic criteria for the ADHD (minimum six symptoms) and also in the children with a smaller number of ADHD symptoms. The ratio of the two fatty acids was associated with the number of the ADHD symptoms present but not with the diagnosis of the disorder, and only in the assessment carried out at seven years of age.

The authors suggest that the assessment carried out at four years of age may have been affected by a measurement error because the ADHD symptoms reported at early ages may be caused by a neurodevelopmental delay falling within the normal range.”Our findings are in line with previous studies that established a relationship between the omega-6: omega-3 ratio in mothers and various early neurodevelopmental outcomes,” commented a researcher and lead author of the study.

“Although the association was not clinically significant, our findings are important at the level of the population as a whole,” noted the author.”If a large proportion of the population is exposed to a high omega-6: omega-3 ratio, the distribution for the ADHD symptom scores would likely move to the right and the prevalence of extreme values would increase, leading to a negative impact on the community`s health costs and productivity,” he added.

“This study adds more evidence to the growing body of research on the importance of maternal diet during pregnancy,” commented the co-author of the study.”The nutrient supply during the earliest stages of life is essential in that it programs the structure and function of the organs, and this programming, in turn, has an impact on health at every stage of life. As the brain takes a long time to develop, it is particularly vulnerable to misprogramming. Alterations of this sort could, therefore, lead to neurodevelopmental disorders.”
