High blood sugar and frailty can be reversed in obesity-related diabetes

Researchers have found in a new study that liver metabolism is disrupted in people with obesity-related type 2 diabetes, which contributes to high blood sugar and muscle loss- also known as skeletal muscle atrophy.

Using human trials as well as mouse models, collaborative research has found the liver metabolism, of the amino acid alanine is altered in people with obesity-related type 2 diabetes. By selectively silencing enzymes that break down alanine in liver cells, high blood sugar and muscle loss can be reversed by the restoration of skeletal muscle protein synthesis, a critical determinant of muscle size and strength.

The findings of the study have revealed that the altered liver metabolism directly affects muscle size and strength and the mechanism behind this is driven by elevated levels of the hormones cortisol and glucagon which enhance the cycling of amino acids between liver and skeletal muscle, causing muscles to become smaller and weaker.

Along with metabolic dysfunction and related complications, as often overlooked co-morbidity of obesity is skeletal muscle atrophy, which causes frailty, and is related to reduced life-quality and death.

The aging-related diseases of skeletal muscle loss and type 2 diabetes are very prevalent and are a huge societal and economic burden. We have known for some time that the aging-related diseases of skeletal muscle loss and type 2 diabetes were linked but we didn’t know how, the Dr. said.

The Dr/ adds, our studies demonstrate that the liver is a critical control point for muscle protein metabolism; a discovery that is quite surprising. We believe that our new findings highlight the need to examine the role of skeletal muscle atrophy in type 2 diabetes more closely in human clinical populations.

The study solidifies the long-known metabolic biochemistry staple, the glucose-alanine cycle, as a fundamental part of metabolism on health and disease.

Biological age of humans reversed by years in groundbreaking study, scientists suggest

Scientists might be able to reverse process of ageing, a new study suggests.

Volunteers who were given a cocktail of drugs for a year actually “aged backwards”, losing an average of 2.5 years from their biological ages, according to the new study. The research showed that the marks on their genomes that represent their " epigenetic clock ", as well as their immune systems, actually improved despite the passing of time.

The scientists involved in the study were shocked by the results. 

“I’d expected to see slowing down of the clock, but not a reversal,” told the researcher who first reported the findings. “That felt kind of futuristic.”

Scientists caution that the study was done with a very limited number of participants: only nine people took the drug cocktail, and there was no control group. But if it is confirmed by further research it could have huge impacts on healthcare, the treatment of disease and how people think about ageing.

In the study, participants were given a growth hormone and two diabetes medications. Scientists then monitored the test subjects’ epigenetic clocks, to understand the effect on how they aged.

The epigenetic clock is measured by the body’s epigenome â€“ a record of chemical changes to an organism’s DNA. As people age, chemical modifications or tags are added to people’s DNA, and those change throughout their lives, so by looking at those tags a person’s biological age can be measured.

Researchers had actually intended to look at how the growth hormone would change the tissue in the thymus gland, which helps with the body’s immune functions and sits in the chest. It normally shrinks after puberty but they hoped to see whether it could be pushed to regrow, by giving participants the growth hormone.

It was only as a secondary consideration that researchers then checked how the drugs changed their epigenetic clocks. The study had finished when the analysis began.


Professor Horvath then looked at four different measures of the epigenetic clock to understand the differing ages of each of the patients. And he found that every one of them had reversed significantly – so significantly that he is optimistic about the results, despite the limited number of participants.

Scientists now hope to test the same effects with more people, through a controlled study, and with different age groups, ethnicities and with women.

The changes could still be seen in the blood of six participants who provided samples long after the study finished.

Some of the drugs used in the cocktail are already being researched as ways of fighting age-related diseases. But the discovery of the combined effect of the three of them could have major implications for the ways that a variety of different drugs are tested, scientists say.

BP pill has potential to slow down Alzheimer's

A high blood pressure drug has the potential to slow down Alzheimer's disease by improving the flow of blood to parts of the brain linked to memory, a study suggests.

Researchers found the drug nilvadipine increased blood flow to the brain's memory and learning centre among people with Alzheimer's without affecting other parts of the brain.


The findings indicate that the known decrease in cerebral blood flow in patients with Alzheimer's can be reversed in some regions.


However, an important question is whether this observed increase in cerebral blood flow translates to clinical benefits, the researchers said.


Alzheimer's disease is the most common form of dementia.


The risk for the disease increases with age and the causes are largely unknown.


Previous research has shown that blood flow to the brain declines in early Alzheimer's disease.
Nilvadipine is a calcium channel blocker used to treat high blood pressure.


Researchers sought to discover whether nilvadipine could help treat Alzheimer's disease by comparing the use of nilvadipine and a placebo among people with mild to moderate Alzheimer's disease.


They randomly assigned 44 participants to receive either nilvadipine or a placebo for 6 months.


Neither researchers nor the participants knew who received the drug or the placebo that was evenly divided among the 2 groups.


At the study's start after 6 months, researchers measured blood flow to specific regions of the brain using a unique magnetic resonance imaging (MRI) technique.


Results showed that blood flow to the hippocampus- the brain's memory and learning center-increased by 20 % among the nilvadipine group compared to the placebo group.


Blood flow to other regions of the brain was unchanged in both groups.


This high BP treatment holds promise as it doesn't appear to decrease blood flow to the brain, which could cause more harm than benefit, said an associate Prof.


Even though no medical treatment is without risk, getting treatment for high BP could be important to maintain brain health in patients with Alzheimer's disease, said the Prof.


Researchers note that sample sizes were too small and follow-up time too short to reliably study the effects of this cerebral blood flow increase on structural brain measures and cognitive measures.


Study participants were screened between 2013 and 2015 as part of a larger research project comparing nilvadipine to placebo among more than 500 people with mild to moderate Alzheimer's disease.


In that larger project, effects on cerebral blood flow were not measured . Overall, no clinical benefit was noted with the use of nilvadipine.


However, a subgroup of patients with only mild symptoms of disease did show benefit, in the sense of a slower decline in memory.


Previous studies have hinted high blood pressure could reduce the risk of developing dementia.


The researchers think that beneficial effects on brain  blood flow could explain part of this effect.

Experts associate negative emotions with cancer

Chinese researchers have revealed the mechanism of how chronic stress promotes breast cancer development, shedding light on future clinical treatment for cancer.

Cancer patients often suffer negative emotions such as anxiety, despair and fear, which are risk factors facilitating tumour growth as well as promoting cancer progression. However, the specific mechanisms of how chronic stress affects cancer development remains unknown yet.


Researchers found that chronic stress might increase epinephrine levels, which enhances lactate dehydrogenase A (LDHA) and promotes breast cancer stem-like cells, it was reported.


Using a drug screen that targeted LDHA, they found that Vitamin C reversed the chronic stress-induced cancer stem-like phenotype.


The study demonstrates the critical importance of psychological factors in promoting stem-like properties in breast cancer cells and provides a promising therapeutic approach for breast cancer, according to a researcher.


The LDHA-lowering agent Vitamin C can be a potential approach for combating stress-associated breast cancer, the researcher said.


This team has been engaged in the dynamic regulation of cancer stem cells research as well as the mechanism of psychological behaviour affecting tumour development.


The researcher noted that patients with breast cancer, ovarian cancer and stomach cancer often have negative emotions, which in turn accelerates the development of their own tumours.


It is necessary to monitor their chronic stress comprehensively by taking psychological assessments as well as conducting blood tests which include epinephrine levels, he said.
