Botanical drug shown to help patients with head and neck cancers in clinical trial

A botanical drug has been shown to help patients with head and neck cancers in a clinical trial. 

In a UCLA-led phase I clinical trial, a new plant-based drug called APG-157 showed signs of helping patients fight oral and oropharyngeal cancers. These cancers are located in the head and the neck. 


Cancers of the head and neck account for 4% of all cancers. About 650,000 new cases are reported each year around the world. People with advanced head and neck cancers have a low survival rate and current treatment options such as surgery, radiation and chemotherapy can have adverse effects. Therefore, more effective and less toxic therapies are needed to help improve the quality of life and outcome for those with these cancers.

APG-157 is a botanical drug developed under the FDA's Botanical Drug Guidance, which includes requirements for the production of plant-based therapies that are marketed as prescription medications. The drug is made up of botanical compounds including curcumin from the Curcuma longa plant, which is commonly referred to as turmeric and is a member of the ginger family. 


Curcumin is one of the medicinally active or therapeutic molecules that has been tested as a possible treatment to help fight multiple cancers because it is an antioxidant that reduces swelling and inflammation. However, there is poor absorption into the bloodstream when curcumin is taken orally. In this study, UCLA researchers found that when APG-157 is taken through oral mucosal absorption, patients have high levels of curcumin circulating in their blood and absorbed by cancer tissues.

APG-157 is made up of multiple compounds produced by plants, including curcumin. UCLA Jonsson Comprehensive Cancer Center researchers found that treatment with this botanical drug resulted in high concentrations of curcumin and its byproducts circulating in the blood and absorbed by tumor tissues within three hours after being taken orally. 


APG-157 reduced the concentration of cytokines -- proteins involved in inflammation -- in the saliva when administered to cancer patients. The therapy also reduced the relative abundance of Bacteroides species, a group of gram-negative bacteria. Gram negative refers to a group of dangerous bacteria that have an outer layer which hides them from the immune system. The relative abundance of gram-negative bacteria compared to the presence of other types of bacteria is correlated with oral cancer.

APG-157 also resulted in the expression of genes that are associated with attracting immune system T cells to the tumor area. This therapy could have a beneficial effect when used in combination with immunotherapy drugs that help immune system T cells recognize and kill tumors. 


The treatment did not have any adverse effects on the study's participants. 


UCLA researchers conducted the study of APG-157 comparing 12 people who had oral and oropharyngeal cancer with a control group of 13 people who did not have cancer. The reason both the people with cancer and without cancer were part of the study was to show that the drug was not toxic to either people with cancer or those without cancer.


 The medication was given each hour for three hours and was delivered as a lozenge that slowly dissolved in the mouth. Blood and saliva samples were collected beforehand -- each of the three hours the medication was administered -- and 24 hours after the last dosage. 


The medication was given to 12 people (some who had cancer and some who did not) and a placebo was given to 13 people. Blood and electrocardiogram tests did not show increased toxicity in the people who took the active medication in comparison with the people who took the placebo, regardless of whether they had cancer or not. 


For the cancer patients who took the medication, there was a decrease in Bacteroides and an increase in T cells in the tumor tissue as compared to cancer patients who took the placebo. Neither the subjects nor the investigators knew whether the drug or a placebo was given when reviewing the blood and saliva test results of the blinded study. 


APG-157 is a botanical drug that has low toxicity. It works effectively to reduce inflammation that contributes to the growth of cancer cells. It also attracts T cells to the tumor micro-environment. 


When used in combination with immunotherapy drugs, APG-157 might have the ability to make the immune system more effective in attacking head and neck cancers. With potential to inhibit the growth of Bacteroides species, APG-157 could also improve cancer therapy through oral microbial changes.

Scientists have found a cancer-fighting chemical in human skin bacteria

Bacteria that commonly live on your skin's surface just might be protecting you from cancer.

'Because each person has many different [staph] strains on their body, we found 6-HAP genes on about 20 percent of the normal population, ' the researcher said. "Still, the revelation that it engages in anti-cancer activity is entirely new".

The findings could lead to a new way to decrease the risk of skin cancer or a less toxic way to treat existing cancer, the lead researcher told.

"It is early to say for sure", the researcher said. The scientists described that a specific bacteria strain of Staphylococcus epidermidis secretes a chemical that eliminates harmful bacteria which have been depicted as responsible for infections. But the specific bacteria in question - a strain called Staphylococcus epidermidis - appears to produce a specific anti-cancer compound known as 6-N-hydroxyaminopurine (6-HAP), the researchers found. The chemical did not inhibit the growth of normal human skin cells, because they possess 6-HAP-deactivating enzymes that were not present on cancerous cells.

One in five Americans will be diagnosed with skin cancer by age 70, according to data from the Centers for Disease Control and Prevention.

Perhaps, the research findings of this study is bit different from the previous research.

They conducted a research in which testing was performed on mice. More than 95 percent of these are non-melanoma skin cancer, which is typically caused by overexposure to the sun's UV rays. 

Those with the bacteria on their skin that did not make 6-HAP had many skin tumors, but mice with the S. epidermidis strain producing 6-HAP did not. It's still unclear if the absence of this bacteria will trigger the growth and development of cancerous tissue.

A researcher has discovered approximately 2 years ago that a type of common skin bacteria protect against cancer because it produces a protein which can protect humans against the oxidative stress caused by free radicals. We further need to understand how these microbiomes play an important role in human biology. Because of this, future studies seeking to exploit the protective potential of skin bacteria "would have to be attempted with high caution", the researcher warned.

However, she added that bacteria tend to have a high mutation rate, and thus a high potential for giving rise to antibiotic resistance.

HEALTH BENEFITS OF GHEE / CLARIFIED BUTTER

Ghee is a common household item found in all Indian kitchens. Mothers very patiently collect the cream/ malai from the milk which is boiled daily, it is stored in the fridge, stirred every other day adding little curd. Once enough malai is collected, it used to be churned by hand to get butter and buttermilk. With the improvement of technologies, either you put the cream in a blender and use whipper to take out butter or use a hand blender to do the same ! I used to love the fresh butter spread on bread and sprinkle little sugar to eat. In Mumbai, this fresh butter used to be spread on sweet buns and sold- called bun maska ! This butter, when melted over medium fire becomes ghee, which is used in making desserts, on top of rice, roti, naan, paranthas and in making sweets and namkeens ( savory snacks).

Health benefits of ghee-
1. Boosts digestion: According to Ayurveda expert, "Ghee is one of the most easily digestible fats used for cooking. Ghee is a natural remedy for balancing the heat element (pitta) of the body. It aids in digestion, prevents constipation and helps in the excretion of toxins. Having half teaspoon of ghee with moong dal khichdi is a great way to detox."

2. Helps in weight loss: Celebrity Nutritionist explains that the type of saturated fat that Desi Ghee has is the short-chain-fatty acid. This helps to burn the stubborn fat in the body while regulating your metabolism.Ghee is higher in conjugated linoleic acid, an unsaturated fatty acid, which causes fat loss.

New drug to beat brain and breast cancers


Researchers have discovered two chemical compounds that effectively stop the growth of brain cancer cells and breast tumours, opening the way for potential new drugs to be developed.

"It is particularly encouraging for brain tumour patients, who do not currently have effective treatment options besides surgery," said James Turkson from University of Hawaii Cancer Centre in the US.

"The targeted treatments are less toxic and therefore will give cancer patients a better quality of life when both compounds are developed as drugs," Turkson said.

In the study published in the journal Cancer Research, the researchers examined compounds that inhibit Stat3, a protein implicated in a variety of cancers that include brain and breast cancers.

In mouse models of brain and breast cancer, the two compounds effectively inhibited tumour growth, the results said.

The two chemical compounds, a hydroxamic acid-based inhibitor (SH5-07), and a benzoic acid-based inhibitor (SH4-54) designed at the UH Cancer Center stopped the growth of brain and breast cancer cells by blocking a certain function of the Stat3 protein.

The two compounds stop the protein from promoting cancer cells to grow, thus stopping the tumours from growing.

"Our results offer preclinical proof of concept for SH5-07 and SH4-54 as candidates for further development as cancer therapeutics," the study said.
