Here's why cold induces tooth pain, hypersensitivity; some remedies for it

A team of researchers from Boston University have uncovered a new function for odontoblasts, the cells that form dentin, the shell beneath the tooth's enamel that encases the soft dental pulp containing nerves and blood vessels.

The results of the study were published in the journal of Science Advances.

"We found that odontoblasts, which support the shape of the tooth, are also responsible for sensing cold," says pathologist Jochen Lennerz, MD, PhD, one of the paper's senior authors and medical director of the Center for Integrated Diagnostics at Massachusetts General Hospital (MGH). "This research contributes a new function to this cell, which is exciting from a basic-science standpoint. But we now also know how to interfere with this cold-sensing function to inhibit dental pain."

Teeth that hurt from exposure to cold can occur for many reasons. Many people have experienced intense pain from cold when they have a hole in a tooth from an untreated cavity, for example. But teeth can also become very sensitive to cold from gum erosion due to aging. Some cancer patients treated with platinum-based chemotherapies have extreme cold sensitivity all over their bodies. "A breeze on the face registers as extreme pain in the teeth, which may even cause some patients to stop therapy," says Lennerz.

Tooth pain has been notoriously difficult to study. A tooth's hardness makes it a challenging tissue to study and inducing tooth pain in humans requires opening the tooth. The team of researchers, therefore, conducted experiments on mice whose molars were drilled under anaesthesia. Mice with dental injuries manifest pain with their behaviour; they drink up to 300 per cent more sugar water than their littermates without dental injuries, for example. In previous research, the team of investigators had discovered TRCP5, a protein encoded by the TRCP5 gene that is expressed in nerves in many parts of the body. Their earlier discovery allowed the researchers to zero in on TRCP5 as a mediator of pain from cold.

By studying genetically altered mice that did not have the TRCP5 gene, the researchers found that the mice with injured teeth did not manifest the increased drinking behaviour and behaved like mice without dental injuries.

"We now have definitive proof that the temperature sensor TRCP5 transmits cold via the odontoblast and triggers nerves to fire, creating pain and cold hypersensitivity," says Lennerz. "This cold sensitivity may be the body's way to protect a damaged tooth from additional injury."

Specifically, in response to cold, the TRCP5 protein opens channels in the membrane of odontoblasts, enabling other molecules, such as calcium, to enter and interact with the cell. If the tooth's pulp is inflamed from a deep cavity, for example, TRCP5 is overabundant, causing increased electrical signaling via the nerves emerging from the root of the tooth and running to the brain, where pain is perceived. When gums recede from ageing, teeth can become hypersensitive because the odontoblasts are sensing cold in a newly exposed region of the tooth. "Most cells and tissues slow their metabolism in the presence of cold, which is why donor organs are put on ice," says Lennerz. "But TRPC5 makes cells more active in cold, and the odontoblasts' ability to sense cold via TRPC5 makes this discovery so exciting."

Lennerz confirmed the presence of the TRPCS protein in extracted human teeth, which was a technical tour de force. "Our teeth aren't meant to be cut into ultra-thin layers so they can be studied under the microscope," says Lennerz, who first had to decalcify the teeth and put them in epoxy resin before slicing them and identifying the TRPC5 channels in the odontoblasts.

The research team also identified a pharmacological target for minimizing tooth sensitivity to cold. For centuries, oil of cloves has been used as a remedy for tooth pain. The active agent in the oil of cloves is eugenol, which happens to block TRCP5. Toothpaste containing eugenol are already on the market, but the findings of this study may lead to more potent applications to treat teeth that are hypersensitive to cold. And there may be novel applications for eugenol, such as treating patients systemically for extreme cold sensitivity from chemotherapy. "I'm excited to see how other researchers will apply our findings," says Lennerz.

Reversing aging of brains cells now possible ?

A recent study has revealed that our brain stem cell becomes less productive as we age because of the increased brain stiffness. The research also found different ways to reverse the aged and unhealthy brain cells with a healthier one.

As our bodies age, muscles, and joints can become stiff, making everyday movement more difficult. The study emphasises the same for our brains, that age-related brain stiffening has a significant impact on the function of brain stem cells.

A multi-disciplinary research team,  studied young and old rat brains to understand the impact of age-related brain stiffening on the function of oligodendrocyte progenitor cells (OPCs).

These cells are a type of brain stem cell important for maintaining normal brain function, and for the regeneration of myelin a fatty sheath that surrounds our nerves, which is damaged in multiple sclerosis (MS). The effects of age on these cells contribute to MS, but their function also declines with age in healthy people.

To determine whether the loss of function in aged OPCs was reversible, the researchers transplanted older OPCs from aged rats into the soft, spongy brains of younger animals. Remarkably, the older brain cells were rejuvenated and began to behave like the younger, more vigorous cells.

The researchers developed new materials in the lab with varying degrees of stiffness and used these to grow and study the rat brain stem cells in a controlled environment. The materials were engineered to have a similar softness to either young or old brains.

To fully understand how brain softness and stiffness influences cell behaviour, the researchers investigated Piezo1 -- a protein found on the cell surface, which informs the cell whether the surrounding environment is soft or stiff.

"We were fascinated to see that when we grew young, functioning rat brain stem cells on the stiff material, the cells became dysfunctional and lost their ability to regenerate, and in fact began to function like aged cells. What was especially interesting, however, was that when the old brain cells were grown on the soft material, they began to function like young cells, in other words, they were rejuvenated," said one of the researchers who carried out the study.

"When we removed Piezo1 from the surface of aged brain stem cells, we were able to trick the cells into perceiving a soft surrounding environment, even when they were growing on the stiff material," explained co-author.

"What's more, we were able to delete Piezo1 in the OPCs within the aged rat brains, which lead to the cells becoming rejuvenated and once again able to assume their normal regenerative function," added the researcher.

Researchers develop 'one-two punch' to destroy cancerous ovarian cells

Researchers have developed a two-step combination therapy 'one-two punch' to destroy cancer cells.

The study showed the superior therapeutic effectiveness of the 'one-two punch' on cells of ovarian cancer patients, based on the manipulation of the state of cellular aging.

"In the case of epithelial ovarian cancer (EOC)--the most common and lethal ovarian cancer--we act in two stages. First, we force the cancer cells to age prematurely i.e., we force them into senescence. This is the first therapeutic punch. We throw our second punch using senolysis, destroying and eliminating them. This strategy requires excellent coordination of the two steps," explained  a researcher.

The team of researchers, discovered that EOC cells enter senescence following chemotherapy in combination with PARP inhibitors. PARPs are enzymes that help repair damage to DNA. By blocking PARPs, PARP inhibitors prevent cancer cells from repairing their DNA, stop them from proliferating and cause them to age prematurely.

"Thanks to our 'one-two punch' approach, we have managed to destroy senescent EOC cells in preclinical ovarian cancer models. Our approach could improve the effectiveness of chemotherapy in combination with PARP inhibitors and counteract the systematic resistance that develops with this treatment," said another researcher.

"Our 'one-two punch strategy' was also tested on preclinical ovarian and breast cancer models, which allowed us to validate its effectiveness," commented Mes-Masson.

Although the results of this study will be used to propose clinical trials for ovarian and triple-negative breast cancer, Rodier said that it is important to remember that they used preclinical models in which there was no immune system. "Given the importance of the immune response in humans, we need to continue evaluating our strategy in a context closer to biological reality."

Everything Is Energy And We Control It With The Power Of Our Thoughts

The Nobel Prize awardees in physics, beyond doubt, proved that the physical world is one big ocean of energy that materializes and dematerializes in a split second, over and over again. Nothing is solid. This is the world of quantum physics.

They proved that thoughts are responsible for holding this ever changing energy field together in the form and shape of the objects we know.

So why do we see a person, instead of flashing clusters of energy?

Think of a film roll. A film is a collection of about 24 frames per second. Each frame is separated from the other by a small space. Compared to the speed these frames change with, our eyes are tricked and see them as a continual film.

Think about television. The cathode tube is a simple tube with a bunch of electrons hitting the screen in a certain way, making an illusion of shape and movement.

Anyway, this is how all objects are made.

We have five physical senses (sight, hearing, touch, smell and taste). Each of these senses has a specific range (for an example, a dog hears a different range of sound than people do; snakes see a different spectrum of light than us, and so on). In other words, our senses perceive energy from a certain fixed point of view and that’s how they create images and form our perception.

But that’s neither exact nor complete. That’s just an INTERPRETATION.

All our interpretations are based exclusively on our “inner map” of reality, but not the real truth. Our “map” is the result of the collective experience of our personal lives.

Our thoughts are associated with this invisible energy and they determine what form it will take. 

They can literally change the universe “particle after particle”, creating our 3D life.

Look around you.

Everything you see in our physical world began as an idea which kept growing until it materialized into a physical object through a series of steps.

You literally turn into your most frequent thoughts.

Your life has become what you imagined and believed in.

The world is literally your mirror, allows you to experience everything you believe to be the truth in this 3D plane … until you change it.

Quantum physics tells us that the world is not a constant as it may appear to be. Instead, it is a place of a constant motion which our individual and collective thoughts keep building, tearing down, and rebuilding. It’s a perpetuum mobile.

What we believe to be true is actually an illusion, almost magic.
 
Fortunately, we started to expose the illusion and, most importantly, we are learning how to change it.
Here’s a simple explanation.

What are the components of our body?

The human body is comprised of nine systems, including the circulatory, digestive, endocrine, muscular, nervous, reproductive, respiratory, skeletal and urinary system.
  1. What makes these systems?
-Tissues and organs.
  1. What are tissues and organs made of?
-Cells.
  1. What makes the cells?
– Molecules.
  1. What makes a molecule?
– Atoms.
  1. What makes an atom?
Sub-atomic particles.
  1. What makes a subatomic particle?
Energy!
We are just an energy light in its most beautiful and intelligent configuration. An energy which is constantly changing under the surface and all of that it’s controlled by our powerful mind.

You are one big and powerful human being.

If you could see under a strong electronic microscope and do other experiments on it, you would see that you are made up of clusters of forever changing energy in the shape of electrons, neutrons, photons and other sub-atomic particles.

So is everything else around you. Quantum physics tells us that the act of observing the object is the one that causes the object to be there.

The object does not exist independently of its observer! So, as you can see, during observation, your attention is focused on something and your intention literally creates that thing.

That is scientifically proven.

Your world is consisted of soul, mind and body.

Each of these three has a function that is unique. What you can see with your own eyes and experience with your own body is the physical world, which we call the BODY. The body is an effect created by a cause.

That cause is called THOUGHT.

The body cannot produce. It can only experience and be experienced … it’s a unique ability. On the other hand, the thought cannot experience … it can make, create and interpret. It needs a world of relativity (the physical world, Body) to be experienced.

The soul is everything, the one that gives life thought and body.

The body has no power to create, although it gives the illusion that it does. This illusion is the reason behind so many frustrations. The body is a pure effect and has no power to cause or create.

The key is to use this information to see the Universe in a different way than we see it now so that we can manifest what we really want.

Adhesive gel to repair eye injury without surgery

 American scientists have designed an adhesive gel that can seal wound or ulcers on the surface of the eye, thus sparing the need for eye surgery.

The study  showed that the gel packed with light-activated chemicals can not only close the defect but also regenerate it, the news agency reported.

“We wanted this material to allow the cells of the cornea to mesh with the adhesive and to regenerate over time to mimic something as close to the native cornea as possible,” said the paper’s co-corresponding author, a professor of ophthalmology.

The gel is clear and viscous in a dropper or syringe, but when exposed to blue light in a short time, it hardens to take on features of a native cornea, and the the cornea cells gradually grow into and become one with the gel, according to the study.

The gel is the first to use visible blue light as opposed to ultraviolet light, which carries a level of toxicity.

In a pre-clinical study, the researchers administered the gel at 20 per cent concentration to corneal defects of 3 mm, and applied visible light for four minutes, leading to a firm adhesion to the defect.

One day later, they observed a transparent, smooth eye surface without inflammation. Over time, the tissue regenerated and the new tissue showed few differences with the native one, according to the study.

The researchers expected to start clinical trials to test the technology in human patients in approximately one year.

Corneal injuries are a common cause of visual impairment worldwide, with more than 1.5 million new cases of corneal blindness reported every year. Some of them require corneal transplants that carry risks of post-transplant complications like infection or rejection.
