Estrogen may facilitate the growth of liver metastases in non-sex-specific cancers

It is known that the tumour microenvironment plays an important role in the progression of cancer. But could estrogen present in this microenvironment facilitate the growth of liver metastases in women affected by colon, pancreatic and lung cancers? This is what a team of researchers from the Research Institute of the McGill University Health Centre (RI-MUHC) suggests, in a study that shows for the first time that the liver immune microenvironment reacts to metastatic cells differently in male and in female mice and that, indeed, the main female hormone estrogen can indirectly contribute to the growth of metastases. Their findings, recently published in Nature Communications, provide a rationale for further exploration of the role of sex hormones in female cancer patients and the potential benefits of anti-estrogen drugs such as tamoxifen in the treatment of hormone-independent cancers that metastasize to the liver.

"What prompted us to begin this investigation was our finding that the immune microenvironments of liver metastases in male and female mice are regulated differently," says Dr. Pnina Brodt, senior scientist in the Cancer Research Program at the RI-MUHC and lead author of the study. "We were also aware of reports based on recent immunotherapy clinical trials that female patients do not respond to immunotherapy as well as male patients and that overall, patients with liver metastases respond more poorly to such treatments than patients without liver metastases."

"We therefore wanted to identify factors unique to male and female responses to liver metastatic disease that could explain these findings and could eventually lead to improved immunotherapy outcomes for patients," adds Dr. Brodt.

Dr. Brodt and her team looked at the interaction between metastatic cells entering the liver and the unique microenvironment of this organ, in mouse models of colon, pancreatic and lung carcinoma, malignant diseases that do not have a clear sex bias and are not depended on sex hormones for growth.

By increasing and decreasing the levels of estrogen in their mouse models, the team discovered that estrogen plays a role in the expansion of liver metastases--a major cause of cancer-associated death. Specifically, they found that this hormone regulates the accumulation in the liver of bone marrow-derived innate immune cells such as the myeloid-derived suppressor cells (or MDSC) and immunosuppressive macrophages.

Cancer cell killers can become friends of the enemy
When metastatic cells enter the liver, innate immune cells are recruited, some from the bone marrow. These cells have the ability to kill cancer cells, but once in the tumour environment, they can also acquire immunosuppressive and tumour promoting functions.

MDSCs, for instance, do not directly affect cancer cell growth, but can do so by changing the microenvironment around the tumour in the liver.

"MDSCs can impede the activity of T lymphocytes, whose role is to kill the cancer cells. In this way, they act to promote--rather than curtail--the growth of metastases," explains Dr. Brodt, who is also Professor in the Departments of Surgery, Oncology and Medicine at McGill University.

The team observed that estrogen induces an immune tolerant environment that helps the tumour cells grow, and that the removal of estrogen reduces the accumulation and activity of MDSCs that, in some way, become "friends" of the enemy.

"The realization that cancer is a community of cells, not just the cancer cells, but also the cells that surround it has become an important part of how the scientific community now views cancer and how it should be treated," says Dr. Brodt.

Towards gender-specific cancer treatment?
In this era of personalized cancer management and with the worldwide effort to develop drugs that target the tumour microenvironment, awareness of the fact that male and female patients may respond differently to such treatments is very important.

"This should now be taken into account to design better, more gender-specific treatment strategies, especially when we know that colorectal cancer incidence, for instance, is increasing in the younger population," says Dr. Brodt.

There are several approved anti-estrogen drugs currently in routine clinical use. However, their use is restricted to patients with hormone-sensitive cancers such as breast cancer.

Further investigation is warranted to determine if estrogen plays the same role in cancer patients, and if low toxicity anti-estrogen drugs such as tamoxifen could be used in conjunction with immunotherapy to neutralize the immunosuppressive environment in patients who have non-sex-specific cancers, are premenopausal and have higher levels of estrogen.

Green tea could help fight tuberculosis

An antioxidant found in the green tea could become key to tackling tuberculosis, a new research has found.

Through laboratory investigations, the research team from Nanyang Technological University, Singapore (NTU) discovered how the prominent compound, known as epigallocatechin gallate (EGCG), can inhibit the growth of a tuberculosis-causing bacteria strain.

The EGCG does so by binding to an enzyme that provides biological energy for cellular activity.
The process results in a dip in the amount of energy the bacteria has for its cellular processes vital for growth and stability, such as cell wall formation.

“Our discovery of the EGCG’s ability to inhibit the growth of M. tuberculosis will allow us to look at how we can improve the potency of this compound in green tea, and other similar compounds, to develop new drugs to tackle this airborne disease,” said study lead researcher.

According to the researchers, cells require energy for vital processes such as cell wall formation to take place. They get their energy from an energy storage molecule made by an enzyme called Adenosine triphosphate (ATP) synthase.

Without energy for essential cellular activity, a cell loses its stability and eventually dies.

To determine the factors affecting the production of ATP synthase, and thus, the amount of energy a bacterial cell has for growth, the research team studied mycobacterium smegmatis and mycobacterium bovis, both of which belong to the same family as M. tuberculosis.
These mycobacterial strains share a similar structural composition.

The team first found that alterations to the genetic code for ATP synthase resulted in an enzyme that produced fewer energy storage molecules in the bacterial cells, slower cell growth, and an altered colony shape.

With this data, the scientists then screened for and found 20 compounds that could possibly bind to ATP synthase and cause the same blocking effect, and then tested them for their efficacy.

Only EGCG, a natural antioxidant that occurs in a large amount in green tea, showed it had the same crucial effect of reducing energy storage molecules in the bacterial cell, the study said.

The research team is now looking at optimising the activity of EGCG for increased efficiency and potency in fighting the tuberculosis bacteria.

Their ultimate goal is to develop a drug cocktail that will tackle multi-drug resistant tuberculosis.

The findings, could pave the way for the creation of novel drugs to combat tuberculosis, one of the most deadly infectious diseases in the world.

Seven Foods That Could Lower Your Risk of Cancer

The development of cancer, in particular, has been shown to be heavily influenced by your diet. It protect people against cancer completely. The term cancer-fighting foods refers to foods that may lower the risk of developing cancer if a person adds them to their diet along with exercise.

In your diet you have to focus to add eating whole, organic foods over processed and pesticide-laden ones. There are certain nutrients that have been shown to be especially good at striking down cancer cells.Following are 7 foods, ingredients that you should take center stage in any diet meant to prevent cancer or support recovery from it.

Quitting smoking improves the prognosis of cancer patients. For patients with some cancers, quitting smoking at the time of diagnosis may reduce the risk of dying.

1.Folate-Rich foods

Low or deficient folate status is associated with increased risk of many cancers and linked in mutations in DNA. Folic acid supplementation and higher serum levels are associated with increased risk of prostate cancer.Studies shown that consuming 400 micro-grams per day can lower the risk of cancer significantly. Folate is a B-complex vitamin that’s so important it is used to fortify many foods that don’t otherwise contain it.
You can add folate in your diet.if you struggle you can have it by supplement. Adding folate rich foods in your each meal is worthy.Great ways to get folate include orange juice, fortified cereals, peanuts, and beans. Spinach and romaine lettuce are also rich in folate, as is asparagus and Brussels sprouts.

2.Vitamin D

Vitamin D is the name given to a group of fat-soluble prohormones required for the regulation of the minerals calcium and phosphorus found in the body. It also plays an important role in maintaining proper bone structure.and helps protect against depression, heart disease, and weight gain.
It also works to curb the growth of cancer cells. In fact, The analysis showed that people who took vitamin D supplements had a 13% lower risk of dying from cancer than those who took a placebo over the same period.
The two main ways to get vitamin D are by exposing your bare skin to sunlight and by taking vitamin D supplements. You can’t get the right amount of vitamin D your body needs from food. The most natural way to get vitamin D is by exposing your bare skin to sunlight (ultraviolet B rays). Salmon.milk, eggs, and seafood such as cod, shrimp, and Chinook salmon contains vitamin D.

3.Tea

Tea is one of the most ancient and popular beverages consumed around the world. Black tea accounts for about 75 percent of the world’s tea consumption.Tea is particularly rich in polyphenols, including catechins, theaflavins and thearubigins.

This comforting and highly customizable beverage is loaded with antioxidants, which are your best line of defense against free radical damage.Tea polyphenols act as antioxidants. When free radicals are allowed to damage cells, it leaves them weakened and vulnerable to cancer. One particular antioxidant in tea, called kaempferol, has been shown in studies to reduce the risk, especially in reproductive cancers.

The Harvard School of Public Health recommends consuming between 10-12 milligrams of kaempferol each day. It takes about four cups to get there. so it is little harder.so you can consume it by diet.Other food sources include fruits like apples, peaches, blackberries, raspberries, grapes, and “technically a fruit” tomatoes. Veggies high in kaempferol are broccoli, Brussels sprouts, potatoes, onions, squash, green beans, cucumbers, lettuce, and spinach.

4.Cruciferous Vegetables

Cruciferous vegetables are vegetables of the family Brassicaceae with many genera, species, and cultivars being raised for food production such as cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts and similar green leaf vegetables. Broccoli and Brussels belong to the group of cruciferous vegetables. Which can reduce the risk of cancer.
There are plenty of ways to incorporate cruciferous vegetables into your diet. The only thing you can’t do (and still get the benefits) is swallow them whole. veggies seem particularly good at protecting against prostate and colon cancers. In fact, in animal studies sulforaphane and other substances produced upon cutting or chewing these veggies actually shrunk tumors.

5. Curcumin

Curcumin known as turmeric is the bright yellow spice that gives curry dishes their distinctive taste and color. It is also known for having anti-inflammatory effects that can fight cancer by suppress the inflammation and prevent the transformation and proliferation of cancer cells.. Inflammation is a factor in the development of pretty much all diseases, and cancer is no exception.
It’s being explored as a cancer treatment in part because inflammation appears to play a role in cancer. Laboratory and animal research suggests that curcumin may prevent cancer, slow the spread of cancer, make chemotherapy more effective and protect healthy cells from damage by radiation therapy.
Perhaps best at reducing the risk of bladder and gastrointestinal cancers, researchers nonetheless insist that no cancer is immune from curcumin’s effects. Dried and powdered curcumin can be sprinkled into all sorts of savory dishes to provide a little Indian flair along with cancer protection.

6. Ginger

Ginger is another popular spice, traditional in Asian cuisine but utilized heavily in Europe and the Americas, too. Ginger is commonly used for various types of “stomach problems,” including motion sickness, morning sickness, colic, upset stomach, gas, diarrhea, irritable bowel syndrome (IBS), nausea, nausea caused by cancer treatment, nausea caused by HIV/AIDS treatment,Beyond being an excellent preventative in our diets, ginger may soon be an integral part of cancer treatments.
Ginger May Have Cancer-Fighting Qualities. Oct. 11, 2011 — Ginger supplements appear to lower some indicators of inflammation in the colon, a new study shows. The study, published in Cancer Prevention Research, is an early step toward finding out whether compounds found in ginger root might prevent colon cancer.Researchers have discovered that ginger can cause the death of cancer cells in two distinct ways. One is called apoptosis, in which the cancer cells basically commit suicide without harming healthy cells that surround them. Another way is autophagy, in which cancer cells are tricked into digesting themselves. Either way, adding more ginger to your diet is a no-brainer.

7. Berries

Berries may be among the most beneficial fruits to eat for cancer prevention.Research is providing new evidence that berries not only contain strong antioxidants that help to prevent cell damage that precedes cancer.they also appear to affect genes that are associated with inflammation and the growth of cancer.
All berries feature high levels of phytonutrients, which are known to offer protection against cancer. Black raspberries could be the cream of the crop though, as they contain a great deal of a particular phytochemical called anthocyanin. Anthocyanin has been found to slow the growth of premalignant cells. It also prevents the growth of new blood vessels that would feed the resulting tumor.
Berries seem to especially protect against colon, esophageal, oral, and skin cancers. Scientific studies typically use concentrated berry powder, but adding a few servings of fresh or frozen berries to your daily diet is also quite beneficial.

Some cancer risk factors, such as genetics and environment, are out of your control, but research suggest that about 70% of your lifetime risk of cancer is within your power to change, including your diet. Avoiding cigarettes, limiting alcohol, reaching a healthy weight, and getting regular exercise are all great steps for preventing cancer. Adopting a healthy diet can also play a vital role.
You can multi-task your cancer prevention diet as well with ginger tea or a veggie omelet sprinkled with curcumin. Since you are starting with fresh, whole foods, the only limit to the diverse meals you can make is your creativity!

Myth about chopped onions causing food poisoning busted

A claim has been afloat on social media for several days now. It says, that leaving chopped up onions overnight can make them toxic, which in turn, can lead to food poisoning. It seems to be doing the internet rounds since 2016. So, is it ideal to immediately cook the onions after you cut them? Read on to find out:

Nutritionist  said, “Yes onions should be cut fresh and cooked right away. However, they don’t turn toxic as their acidic pH inhibits microbes and bacterial growth.” She warned that keeping cut onions outside refrigerator can kill their freshness, and it’s not a good idea to consume them the next day.

The claim has swelled on the internet, with the National Onion Association in the US releasing a statement to quell it. They mention, “When cut, onions release compounds that do not promote pathogen growth.” In simple terms, cut onions don’t necessarily lead to disease formations. “When handled properly, cut onions can be stored in the refrigerator in a sealed container for up to 7 days,” it read.

Always go for fresh ingredients
Nonetheless, going for fresh ingredients is always a better option. Expert states, “Juice released from cut onion is known to kill or inhibit the growth of several types of microorganisms, including some of those capable of causing food poisoning in humans.”

Myth busting site explained in 2009 that leaving cut onion for a few days doesn’t let the onions get contaminated and don’t cause food poisoning, thereby busting the myth. But, it’s ideal to use a fresh knife when you’re cutting or chopping anything, and make sure everything has been washed thoroughly.  “It is a safe and hygienic practice to use freshly cut vegetables- and the same applies to onions too,” the nutritionist said.

The nutritionist added, “Ideally even leftover salad should not be stored but eaten immediately.” For cooking too, make use of fresh ingredients, to extract the maximum health benefits.

Cutting an onion and keeping it near you in a small container for 5 days at the very first sign of cold or fever, will protect you from suffering from them. As, onion takes the bacteria present around and it turns black or dark brown if there are more bacteria around you. I've tried it and it has worked.

Coffee compounds may help to reduce prostate cancer risk

Scientists have identified kahweol acetate and cafestol hydrocarbon compounds naturally found in Arabica coffee which may inhibit the growth of prostate cancer.

The pilot study showed kahweol acetate and cafestol can inhibit growth in cells that are resistant to common anti-cancer drugs like Cabazitaxel.

“We found that kahweol acetate and cafestol inhibited the growth of cancer cells in mice, but the combination seemed to work synergistically, leading to a significantly slower tumour growth than in untreated mice,” said lead author.

For the study, presented at the European Association of Urology Congress in Barcelona, the team tested six compounds, naturally found in coffee, on proliferation of human prostate cancers cells in vitro (i.e. in a petri-dish).

They found cells treated with kahweol acetate and cafestol grew more slowly than controls. They then tested these compounds on prostate cancer cells, transplanted to mice (16 mice).

“After 11 days, the untreated tumours had grown by around three and a half times the original volume (342 per cent), whereas tumours in the mice treated with both compounds had grown by just over one and a half (167 per cent) times the original size,” he said.

It also showed the growth reduction occurred in transplanted tumour cells, rather than in native tumour cells.

Although “these are promising findings, but they should not make people change their coffee consumption,” cautioned Professor from the varsity.

“Coffee can have both positive and negative effects. We need to find out more about the mechanisms behind these findings before we can think about clinical applications. But if we can confirm these results, we may have candidates to treat drug-resistant prostate cancer,” he noted.
