Novel saliva test can detect mouth, throat cancer early

A non-invasive saliva test can detect human papilloma virus-16, the strain associated with oropharyngeal cancer (OPC), showing promise for earlier and easier detection of mouth and throat cancer, report researchers.

The novel technique detected OPC in whole saliva in 40 per cent of patients tested and 80 per cent of confirmed OPC patients. OPC has an approximate incidence of 115,000 cases per year worldwide and is one of the fastest-rising cancers owing to increasing HPV-related incidence, especially in younger patients.

“It is paramount that surveillance methods are developed to improve early detection and outcomes,” said co-lead investigator. Cancers that occur in the back of the mouth and upper throat are often not diagnosed until they become advanced, partly because their location makes them difficult to see during routine clinical exams.

“The successful detection of HPV from salivary exosomes isolated by our acoustofluidic platform offers distinct advantages, including early detection, risk assessment and screening,” added Dr Huang in a paper published in The Journal of Molecular Diagnostics. This technique may also help physicians predict which patients will respond well to radiation therapy or achieve longer progression-free survival.

In the study, investigators analyzed saliva samples from 10 patients diagnosed with HPV-OPC using traditional methods. They found that the technique identified the tumour biomarker in 80 per cent of the cases when coupled with the traditional detection method called droplet digital PCR. “The saliva exosome liquid biopsy is an effective early detection and risk assessment approach for OPC,” said co-lead investigator.

According to the researchers, this technology can also be used to analyze other biofluids such as blood, urine and plasma.

Laser-based saliva test to help diagnose diseases early

A novel laser-based technique can analyse human saliva and non-invasively diagnose immune system disorders even before the symptoms appear, increasing the chances of recovery, scientists say.

Disorders of the immune system lead to various diseases from allergy and type 1 diabetes to more dangerous autoimmune diseases, such as multiple sclerosis

However, such  diseases are diagnosed only when the symptoms are clearly demonstrated.
Researchers developed a unique method of immune diseases diagnosing before the symptoms appear.
They proposed a laser-correlation spectroscopic technique (also called dynamic light scattering) for studying the immune response in body fluids, for example, in saliva.
"We offer a cheap and effective method of diagnosing the diseases without any indication. We need a laser, a receiver and a programme for data analysis, she said.

In the course of routine clinical examination, saliva can be tested so that the patient can learn more about the disease before the symptoms appear. This method will increase the chances of recovery," she said.

It is known that saliva contains the same immune proteins as blood. Moreover, it is much easier and cheaper in comparison with blood analysis.

The proposed method is to analyse the scattered light obtained by laser illumination of human's biological fluid (saliva or blood).

The laser beam is focused on the sample. The proteins in the liquid scatter the light, which is registered by the detector.

By analysing the change in the intensity of scattered light in time, it is possible to determine the size of the particles floating in the liquid, researchers said.

The particle size changes during the activation of immunity, the proteins are binding and become larger.

In addition, the size composition of biological fluids in different people can vary. It depends on the certain diseases in the organism.

Thus, it is possible to determine whether the human body reacts to infections properly and to diagnose the diseases.

With this method, it is also possible to test medical drugs not on a human, but on his biological fluids, paving the way for custom treatments to suit the individual.

Such studies of the dynamics of the activation of immune proteins using optical methods have not yet been carried out, thus the research remains to be done.

The current task is to create a detailed study of the reactions of donors with various pathologies.

Health Tests All Men Over 40 Should Undergo

As you start getting older, it's essential to start getting yourself medically tested on a regular basis, in order to keep yourself as strong and healthy as can possibly be. That's why we've compiled a list of tests that every man over 40 should start taking.

1. Testosterone
Low levels of testosterone may cause many unpleasant changes in your body, such as fatigue, erectile dysfunction, weight gain , loss of body hair, loss of muscle mass , difficulty concentrating, trouble sleeping, a reduction in bone density, or even subtle changes in your personality. Your doctor can quickly assess your testosterone levels through a saliva or blood test.
2. Digital Rectal Exam (DRE)
Undergoing a DRE will help with the early detection of prostate cancer and other irregularities of the prostate gland. Your doctor will need to insert a lubricated gloved finger into your rectum to feel for abnormalities, and while this might sound uncomfortable, it's very important to get it done, because it might just save your life!
3. Blood Pressure
Blood pressure is a symptomless killer, which can wreak havoc on your lungs, brain, heart, kidneys and blood vessels. That's why it is a good idea for over-40s to keep portable blood pressure monitors on them at all times, since there's nothing better than having peace of mind at all times.
4. Prostate-Specific Antigen (PSA)
Taking the PSA test  will help your doctor determine how much prostate-specific antigen, a prostate health indicator, is present in your blood. High or rising levels of PSA could be indicative of prostate cancer or other prostate-related illnesses.
5. Cholesterol
Once you reach the age of 40, you really should get all of your cholesterol levels  checked by your doctor.
These include:

1. Total cholesterol level
2. Low-density lipoprotein (LDL)
3. High-density lipoprotein (HDL)
4. Triglycerides

6. Blood Sugar
This is an important test to help screen for insulin resistance, diabetes, and pre-diabetes. Insulin resistance may cause weight gain, high cholesterol levels, high blood pressure, bloating, or even high blood sugar. If left untreated, diabetes may cause problems with your feet, eyes, skin, heart, kidneys, nerves, and even your mental health.
7. Colorectal Cancer
Doctors recommend that people over 40 get screened for colon cancer with any of the following tests: sigmoidoscopy, colonoscopy, or a fecal occult blood test. A colonoscopy is considered the best option for the prevention of colorectal cancer, but a fecal occult blood test is the best at detecting the early stage of this cancer.

8. HIV
Because birth control becomes less of a concern for people over 40, many of them are having unprotected sex. That's why around a quarter of HIV cases are actually people over 40, so you should still get yourself checked no matter how old you are.

Now a saliva test to detect dengue in 20 minutes

Here's one potent weapon in your fight against Dengue. A team of researchers in Singapore has developed a paper-based disposable device that will allow dengue-specific antibodies to be easily detected from saliva within 20 minutes.

The rapid diagnostic kit can detect a key dengue antibody from saliva that is present in early-stage secondary infection, said researchers from the Institute of Bioengineering and Nanotechnology (IBN).

"The ability to differentiate between primary and secondary dengue infections makes it a valuable early diagnosis tool that would help to ensure timely treatment and proper care of patients," explained professor Jackie Y. Ying, executive director of IBN.

Currently, dengue infection is diagnosed in the laboratory by testing the patient's blood sample for the presence of dengue antigens or antibodies.

The new device is capable of detecting IgG, a dengue-specific antibody found at the onset of secondary infections, directly from saliva in one step. To make the device, the IBN researchers used an innovative stacking flow design to overcome key challenges faced by existing lateral flow designs such as those used in pregnancy test kits.

Patients with secondary infection, who have previously been infected with other serotypes of dengue virus, stand a higher risk of developing dengue hemorrhagic fever or dengue shock syndrome.

The diagnostic kit can also be adapted to detect other infectious diseases such as HIV and syphilis, the researchers noted. They are also investigating the use of other common fluid samples, such as blood, urine and serum for rapid, high-sensitivity test kits.

The results are detailed in the journal Lab on a Chip.
