Researchers Successfully Turn Breast Cancer Cells Into Fat to Stop Them From Spreading

Researchers have been able to coax human breast cancer cells to turn into fat cells in a new proof-of-concept study in mice.

To achieve this feat, the team exploited a weird pathway that metastasising cancer cells have; their results are just a first step, but it's a truly promising approach.

When you cut your finger, or when a foetus grows organs, the epithelium cells begin to look less like themselves, and more 'fluid' – changing into a type of stem cell called a mesenchyme and then reforming into whatever cells the body needs.

This process is called epithelial-mesenchymal transition (EMT) and it's been known for a while that cancer can use both this one and the opposite pathway called MET (mesenchymal‐to‐epithelial transition), to spread throughout the body and metastasise.

The researchers took mice implanted with an aggressive form of human breast cancer, and treated them with both a diabetic drug called rosiglitazone and a cancer treatment called trametinib.

Thanks to these drugs, when cancer cells used one of the above-mentioned transition pathways, instead of spreading they changed from cancer into fat cells – a process called adipogenesis.

"The models used in this study have allowed the evaluation of disseminating cancer cell adipogenesis in the immediate tumour surroundings," the team writes in their paper.

"The results indicate that in a patient-relevant setting combined therapy with rosiglitazone and trametinib specifically targets cancer cells with increased plasticity and induces their adipogenesis."

Although not every cancer cell changed into a fat cell, the ones that underwent adipogenesis didn't change back.

"The breast cancer cells that underwent an EMT not only differentiated into fat cells, but also completely stopped proliferating," says senior author of the study.

"As far as we can tell from long-term culture experiments, the cancer cells-turned-fat cells remain fat cells and do not revert back to breast cancer cells."

So how does this work? Well, as a drug trametinib both increases the transition process of cells - such as cancer cells turning into stem cells - and then increases the conversion of those stem cells into fat cells.

Rosiglitazone was less important, but in combination with trametinib, it also helped the stem cells convert into fat cells.

"Adipogenic differentiation therapy with a combination of rosiglitazone and [trametinib] efficiently inhibits cancer cell invasion, dissemination, and metastasis formation in various preclinical mouse models of breast cancer," the team said.

What's exciting is that these two drugs are already FDA-approved, so it should be easier to get this type of treatment into clinical trials for actual people.
 
That's exciting even despite the fact that we know many mouse-tested treatments don't actually make it to, or fail, the clinical trial stage. The fact this worked on human cancer cells gives a little extra hope.

In the meantime, the team is investigating whether this therapy would work combined with chemotherapy, and whether it would apply to other types of cancers.

"In future, this innovative therapeutic approach could be used in combination with conventional chemotherapy to suppress both primary tumour growth and the formation of deadly metastases," the author said.

"The clinical evaluation of the treatment's repressive effect on experimental breast cancer metastasis and, thus, of its potential in treating stage IV breast cancer will require adjuvant combinations with chemotherapy in advanced preclinical models," the researchers said.

"Since we have used FDA-approved drugs to study the preclinical effect of the treatment, a clinical translation may be possible."

Flu Treatments Explained

The government has just approved a new form of antiviral medication called baloxavir marboxil (Xofluza) for people aged 12 and over. The drug is a single-dose oral medication and works by preventing the flu virus from spreading all over your body. Its creator, claims that it fights a whole range of flu viruses, including those resistant to other flu treatments and to bird flu strains.

Oseltamivir is available both as a generic version or under the trade name of Tamiflu. The latter was initially approved way back in 1999, and the government eventually approved a generic version in 2016. It's available in liquid suspension or pill form and is typically prescribed twice a day for 5 days. It can also be prescribed for flu prevention, in which case it’s taken once daily for seven days.

Zanamivir (Relenza) is a powder that’s inhaled, and the FDA has claimed that it is safe for children 7 years old and above, and is normally taken twice daily for five days. Finally, peramivir (Rapivab) is given intravenously, is safe for children aged two and over, and is administered once time for 15 to 30 minutes.
Who Are These Drugs For?
The people who should most strongly consider taking one of these new medications are:
• Hospitalized flu patients
• People who already have the flu
• People over 65
• Children under 5
• Pregnant women
• New mothers

• People suffering from asthma or organ diseases
• Obese people
• Diabetics
• People with weakened immune systems, such as from cancer, AIDS or HIV

What Side Effects Are There?
Patients on these antivirals typically report nausea as side effects. Other side effects may include headaches and common cold symptoms. When it comes to kids, Tamiflu has been connected to some psychiatric side effects. These could include irritability and occasional seizures. However, these symptoms normally go away when the drug is stopped.
How Effective Are They?
Your mileage may vary, but most professionals say that the drugs shorten the flu by around a day or so. The government approval of Xofluza was based on studies which compared it to a placebo and with oseltamivir (Tamiflu). In a particular study, which included over 1,000 patients, the median time to symptom improvement was around 54 hours with Xofluza and 80 with a placebo.
What Alternatives Are There?
Many people swear by home remedies, such as putting onions in your socks or consuming elderberry. Proponents claim that the onion's acidic compounds kill the nasty flu virus, while elderberry proponents claim that it eases symptoms. However, "there is currently no strong scientific evidence that any natural product is useful against the flu," according to the doctors.

"In terms of natural products, I don't give a recommendation for them," says a physician. However, he also notes that he understands that ''people often feel better doing something than nothing."

This is why the only alternative treatment that healthcare professionals usually provide are fever reducers such as acetaminophen, bed rest, and plenty of fluids.

Hydration is crucially important, according to a professor of infectious diseases and preventive medicine. "You frequently are so ill you have to rouse yourself from your bed of thorns and make yourself drink fluids. Water is fine, flavored or sparkling. Alcohol doesn't count. Coffee and tea don't count, they have caffeine [and are dehydrating]." This is a particularly important point since the Dr. says that "dehydration predisposes you to pneumonia," which is the last thing that someone with the flu needs.

Immunotherapy Drugs Can Melt Away Lung Cancer

Drugs for immune therapy are able to give patients with lung cancer a new chance to enjoy life for some extra years, said doctors on Monday.

They found that patients with lung cancer can be treated with immune therapy drugs before getting surgery to remove the tumor. These drugs will literally melt away the tumor, and they will stop it from spreading.

Other lung cancer patients tried this treatment instead of chemotherapy. A lung cancer specialist, who wasn’t involved in the study said that the results from immunotherapy are startling: “I have never seen progress move so fast. The results today are really a paradigm shift. They will mean more lung cancer patients getting immunotherapy up front.”

Immunotherapy Helping the Body Fight Cancer
Through immunotherapy, the immune system of the body gets help in fighting off cancer: it boosts the immune system activity, it tracks tumor cells and targets tumors.


The treatment includes a new class of drugs that have helped to stall the melanoma of a pateint.
 
And according to the new results shown in a study released, these drugs could change the way lung cancer is treated. 

A team at a medical school has treated 20 of their patients with immune therapy drugs before getting them into surgery to remove the tumors. After one year, 16 patients were alive, with no traces of cancer detectable. Two of them had their cancer back, but after receiving extra treatment, they showed no other symptoms. One of them died of cancer and the other died of a head injury (which had no connection to cancer).

Even though it was a small study, it shows that immunotherapy drugs are a good option before surgery. All these drugs were used on patients with advanced cancer because the drugs are new. There are more drugs available, and for this study, it was used Opdivo, which broke down the tumor for the immune system to fight it easier.

The Good News and The Bad News
The Dr. said that “when the surgeons open the patients, they see the tumor almost melting away. That is extraordinary. It eats it away like a Pac-man. It probably creates a roller coaster of immune response. It is amazing news for patients with this disease.”

The bad news is that not all patients respond to the treatment and it cost over $100,000 – only a course of treatment. The leader of the study, said that he will not use “use the term cure until you survive for 10 years”.
