Goodbye Ozempic? This Experimental Drug Could Burn Fat Without Muscle Loss
The next big name in weight-loss medicine may not be another appetite suppressant. A new experimental compound called TOFA is attracting attention after researchers found that it helped obese mice lose body fat while preserving lean muscle mass, without making them eat less.
The findings are particularly interesting at a time when drugs such as Ozempic and Wegovy have changed obesity treatment. However, the race is now shifting towards an important question: can people lose fat without losing muscle along with it?
Published in Science Advances, the new study suggests TOFA, short for 5-tetradecyloxy-2-furoic acid, may offer a completely different approach to weight loss. However, there is one major catch. So far, the compound has only been tested in animals, not humans.
What is TOFA and How Does it Work?
TOFA, short for 5-tetradecyloxy-2-furoic acid, is an oral compound first studied decades ago as an inhibitor of enzymes involved in making fats. Researchers at the University of California, Berkeley, have now shown that it does more than block lipid production. It also activates cellular receptors that switch on genes helping cells take up and burn fat for energy.
In experiments published in the journal Science Advances, obese mice given TOFA twice daily for four weeks lost an average of 18 percent of their body weight. Food intake stayed the same, physical activity did not increase, and body temperature remained unchanged. Yet whole-body energy expenditure rose by as much as 18 percent. Most of the weight lost came from fat, with little to no reduction in lean muscle mass.
Ozempic vs TOFA: What’s Different?
Current GLP-1 medications such as semaglutide (Ozempic, Wegovy) and tirzepatide (Mounjaro, Zepbound) primarily work by reducing appetite and lowering calorie intake. While highly effective for weight loss, they can sometimes lead to loss of muscle along with fat, and stopping the drugs often brings weight regain.
TOFA targets the other side of the energy balance equation. Instead of making people or animals eat less, it increases the amount of energy the body burns. Senior researcher Anders Näär has described body weight as responding to two levers: taking in fewer calories or spending more energy. GLP-1 drugs mainly pull the first lever; TOFA aims at the second.
This is where the findings become particularly intriguing. In the animal experiments, TOFA-treated mice lost significant body weight and fat mass without a statistically significant reduction in lean muscle mass.
Researchers also found improvements in fasting blood glucose, insulin levels and insulin sensitivity. The compound showed potential benefits for abnormal triglyceride levels and fatty liver-related disease too, suggesting that its effects may go beyond the weighing scale.
What Other Benefits Does TOFA Offer?
Beyond fat loss, treated mice showed improved insulin sensitivity and better blood sugar control. Triglyceride levels dropped, and signs of fatty liver disease improved, including reduced liver fat, inflammation and fibrosis. These metabolic gains occurred without the rise in blood triglycerides that has limited some other drugs in the same chemical class.
When researchers combined lower doses of TOFA with semaglutide or tirzepatide, the results were stronger than either treatment alone. The pairs produced greater reductions in body weight, better glucose control, lower insulin levels and improved triglyceride readings. Scientists view TOFA as potentially complementary rather than a direct replacement for existing medicines.
The findings so far come only from animal studies. Safety, optimal dosing and effectiveness in humans have not yet been established. Clinical trials would be needed before any conclusions can be drawn about its use in people. Researchers have noted that the compound’s dual action on lipid synthesis and energy expenditure appears distinctive, but translation from mice to humans is never guaranteed.
So, goodbye Ozempic? Not quite. But hello to a new possibility. If TOFA’s effects translate to humans, future obesity treatment may focus not only on eating less, but also on helping the body burn more energy while protecting the muscle that people need to stay strong.
